An Efficient Method for Generation of Transgenic Rats Avoiding Embryo Manipulation

Abstract: Although rats are preferred over mice as an animal model, transgenic animals are generated predominantly using mouse embryos. There are limitations in the generation of transgenic rat by embryo manipulation. Unlike mouse embryos, most of the rat embryos do not survive after male pronuclear DNA injection which reduces the efficiency of generation of transgenic rat by this method. More importantly, this method requires hundreds of eggs collected by killing several females for insertion of transgene to generate t… Show more

“…Primer details were given in Supplementary Table S1. The construct was linearized with Stu I , and was used to generate transgenic rat (Tg) as reported by us previously 47 . Briefly, the linearized construct (a total of 30 µg DNA with a concentration of 1 µg/µl) was injected into the testis of 38 days old Wistar rat, and electroporated using 8 square 90 V electric pulses in alternating direction with a time constant of 0.05 second and an inter-pulse interval of ~1 second via an electric pulse generator.…”

“…A large number of murine models are commercially available, and distributers can easily be found on the website of the “International Mouse Strain Resource” (IMSR, ). Even though transgenesis is more complicated in the rat than in the mouse, technical advances have recently been made and, as a result, the number of transgenic rat models for human disease will certainly significantly increase in the coming years [ 146 ].…”

“…Stable anaesthesia is generally easier to achieve, and appropriate monitoring of physiological parameters less invasive in rats due to their larger body size (compared to mice). As transgenic rats are becoming more accessible (Pradhan and Majumdar, 2016), implications of species selection on the expected data quality may become increasingly important for assessing the construct validity of a given experiment.…”
