An Efficient Greener Approach for N-acylation of Amines in Water Using Benzotriazole Chemistry

Abstract: A straightforward, mild and cost-efficient synthesis of various arylamides in water was accomplished using versatile benzotriazole chemistry. Acylation of various amines was achieved in water at room temperature as well as under microwave irradiation. The developed protocol unfolds the synthesis of amino acid aryl amides, drug conjugates and benzimidazoles. The environmentally friendly synthesis, short reaction time, simple workup, high yields, mild conditions and free of racemization are the key advantages of… Show more

“…While acyl-amino acids are environmentally benign regarding application and biological degradation, the conventional chemical synthesis of these compounds via the Schotten-Baumann reaction is not sustainable (Ibrahim et al 2020 ). Chlorinated fatty acids have to be used which are typically produced with toxic phosgene or thionyl chloride.…”

“…Besides using of classical acids and bases, several Lewis acid catalysts, solid acid and heteropolyacid catalysts, organocatalysts, ionic liquids and others have been used for the acylation reactions [21]. Amides are commonly produced by N-acylation of amines [22][23][24].…”
