An easy and efficient inducible CRISPR/Cas9 platform with improved specificity for multiple gene targeting

Abstract: The CRISPR/Cas9 system is a powerful genome editing tool and has been widely used for biomedical research. However, many challenges, such as off-target effects and lack of easy solutions for multiplex targeting, are still limiting its applications. To overcome these challenges, we first developed a highly efficient doxycycline-inducible Cas9-EGFP vector. This vector allowed us to track the cells for uniform temporal control and efficient gene disruption, even in a polyclonal setting. Furthermore, the inducible… Show more

“…as well as the doxycycline-inducible Cas9 viral vectors. 7 Indeed, considerably decreased off-target effects without influence on on-target efficiency have been observed using these systems, a finding that could support the working strategy that Cas9 expression duration can be manipulated to achieve higher specificity. However, there is potential toxicity associated with small molecular inducers.…”

“…Physicians and patients alike are certainly right to seek to the medical risks of vaccines, such as allergies to certain ingredients like eggs or latex,65 and the rarer risks of anaphylaxis and encephalopathy;51 likewise, they must consider the predominant medical risk of CRISPR––off-target effects66 67––which scientists are working to diminish 68. However, to allow fear of these risks to obfuscate these innovations' very tangible benefits would be an irresponsible and potentially more dangerous option.…”
