An arginine/ornithine exchange system in Spiroplasma melliferum

Abstract: University-H ad assa h Medical School, POB 12272, Jerusalem 91 120, Israel Spiroplasma me//ifemm cells utilize arginine via the arginine dihydrolase pathway. L-Arginine uptake by intact cells was a saturable process both as a function of time and arginine concentration (Km = 40 pM). Uptake was not affected by pH in the range pH 59-89, or by Atrulline, o-arginine, L-histidine or L-canavanine at concentrations tenfold higher than that of L-arginine. In contrast, L-arginine uptake was markedly inhibited by L-omit… Show more

“…Spiroplasma melliferum and Sp. floricola were grown in a modified Saglio medium (Shirazi et al, 1995). The cultures were inoculated with a frozen inoculum at a level of 1-5 ' / o and then grown wuthout aeration for 24-48 h. The Mycoplasma and Acholeplasma species were grown at 37 "C whereas the Spiroplasma species were grown at 32 "C. Growth was monitored by measuring the OD,,, of the culture and by recording pH changes in the growth medium.…”

“…There are other arginine-and ornithine-related observations that may be relevant. Arginine/ornithine exchange systems have been described in various bacteria including Spiroplasma melliferum, lactic acid bacteria and P. aeruginosa (see Shirazi et al, 1995). Also, a staphylococcal siderophore, staphyloferrin A, has D-ornithine as a major structural component and addition of D-ornithine to iron-deficient medium stimulates siderophore production markedly (Konetschny-Rapp et al 1990).…”

“…Does the transporter function as uniporter, antiporter, or symporter? Among mollicutes, an arginine-ornithine exchange system has been reported only for spiroplasmas (30). Attempts have also failed to detect an arginine-ornithine exchange system for M. pneumoniae (A. Katzenell, unpublished data), although such a system has been identified in other bacteria, for example, Pseudomonas aeruginosa (38) and Streptococcus lactis (39).…”
