An apple CIPK protein kinase targets a novel residue of AREB transcription factor for ABA‐dependent phosphorylation

Abstract: Phytohormone abscisic acid (ABA) regulates many important processes in plants. It is a major molecule facilitating signal transduction during the abiotic stress response. In this study, an ABA-inducible transcription factor gene, MdAREB2, was identified in apple. Transgenic analysis was performed to characterize its function in ABA sensitivity. Overexpression of the MdAREB2 gene increased ABA sensitivity in the transgenic apple compared with the wild-type (WT) control. In addition, it was found that the protei… Show more

“…Sixty-eight MdDREB genes have been classified into six subgroups against the entire genome of apple, of which MdDREB2A and MdDREB2B (called MDP0000147009 and MDP0000153866 in their study) were identified and found they could be up-regulated under various abiotic stress treatments (Zhao et al, 2012). MdAREB1A and MdAREB1B (called MdAREB3.2 and MdAREB2 in their study) were identified and studied for their sensitivity to ABA in apple (Ma et al, 2017). MdRD22, MdRD29A and MdRD29B were also found to be responsive for drought or osmotic stress Shao et al, 2019).…”

“…Our yeast two hybrid experiment showed that there was interaction between FtbZIP5 and the homologous protein FtSnRK2.6 of SnRK2.6, but their phosphorylation requires further verification. Similarly, in apple, MdAREB2 is phosphorylated by the SnRK2 homologous protein MdCIPK22 [46]. Previous studies have found an interaction between FtbZIP83 and FtSnRK2.6/2.3 [23].…”

“…For CO‐IP analysis, precipitates were further analysed by SDS‐PAGE and Western Blotting as described by Ma et al . (). The proteins from crude lysates (input) were detected with anti‐Myc antibody, and the IPed proteins (output) were detected with anti‐HA antibody.…”

“…After brief centrifugation, four washing steps followed, after which loading buffer was added to the precipitates and boiled. For CO-IP analysis, precipitates were further analysed by SDS-PAGE and Western Blotting as described by Ma et al (2017b). The proteins from crude lysates (input) were detected with anti-Myc antibody, and the IPed proteins (output) were detected with anti-HA antibody.…”
