An antiproliferative BMP-2/PPARγ/apoE axis in human and murine SMCs and its role in pulmonary hypertension

Abstract: Loss-of-function mutations in bone morphogenetic protein receptor II (BMP-RII) are linked to pulmonary arterial hypertension (PAH); the ligand for BMP-RII, BMP-2, is a negative regulator of SMC growth. Here, we report an interplay between PPARγ and its transcriptional target apoE downstream of BMP-2 signaling. BMP-2/BMP-RII signaling prevented PDGF-BB-induced proliferation of human and murine pulmonary artery SMCs (PASMCs) by decreasing nuclear phospho-ERK and inducing DNA binding of PPARγ that is independent … Show more

“…HPAECs were seeded onto 10cm tissue culture dishes at 5x10 Furthermore, vascular cells from IPAH patients are known to maintain a proliferative phenotype in vitro [31][32][33]. These results are consistent with previous findings demonstrating that IL-33 expression is known to be inversely related to cellular proliferation.…”

“…An indicator of RV contractility, RV dP/dt max measured during RV catheterization was significantly increased after 3 weeks of hypoxia in SuHx mice compared with both baseline and VehHx controls and increased in parallel with RVSP and hypertrophy. The latter phenomenon has been observed in several mouse PH studies 10,12,14 and represents increased contractility in the hypertensive and hypertrophic RV. 14 RV dP/dt max returned to normoxic values after 10 weeks of recovery from chronic hypoxia (Fig.…”

“…In addition to eNOS, recent studies demonstrate the involvement of Wnt/␤-catenin signaling in BMP-2 stimulated PAEC migration, which in the healthy pulmonary endothelium is mediated by BMPRII (28,29). Although there are few studies examining cross-talk between NO and Wnt/␤-catenin signaling, there is some evidence of it in the literature.…”
