An androgen receptor negatively induced long non-coding RNA ARNILA binding to miR-204 promotes the invasion and metastasis of triple-negative breast cancer

Abstract: Androgen receptor (AR) is emerging as a novel prognostic biomarker in triple-negative breast cancer (TNBC), but the underlying mechanisms remain unknown. As accumulating evidence has shown that long non-coding RNAs (lncRNAs) regulate important cancer hallmarks, we hypothesised that AR-regulated lncRNAs might play roles in TNBC progression. Here, we performed experiments with or without DHT treatment in three TNBC cell lines, and we identified an AR negatively induced lncRNA (ARNILA), which correlated with poor… Show more

“…Immunohistochemistry. Immunohistochemical staining was performed as previously described (24). Staining was scored as: i) ++++ if >80% tumor cells were immuno-positive; ii) +++ for 51-80%; iii) ++ for 11-50%; iv) + for 1-10% cells; and v) -if <1% of the tumor cells were positively stained.…”

“…Being down‐regulated, miR‐129‐5p is associated with high cell proliferation, migration, and invasionAR negatively induced lncRNA (ARNILA) up‐regulation was related to poor survival in TNBC patients and promoted EMT, invasion and metastasis by sponging miR‐204. One target of ARNILA is SOX4 which is known to induce EMT and contribute to BC progression . GAS5, which is down‐regulated, can suppress TNBC progression by competitively binding miR‐196a‐5p which is targeted to FOXO1/PI3K/AKT signal pathway …”

“…Long noncoding RNA (lncRNAs) and circular RNAs (cir-cRNAs) are important factors in human cancer pathogenesis [15,20,21]. Both lncRNAs and circRNAs lack the ability to encode proteins and circRNAs are characterized by the covalent conjunction and lacking of the 3′ and 5′ end.…”
