An analysis of splicing, actin-binding properties, heterodimerization and molecular interactions of the non-muscle α-actinins

Abstract: The non-muscle α-actinin isoforms (actinin-1 and -4) are closely related dimeric actin filament cross-linking proteins. Despite high sequence similarity, unique properties have been ascribed to actinin-4 in particular. For example, actinin-4, but not actinin-1, is essential for normal glomerular function in the kidney, is overexpressed in several cancers and can translocate to the nucleus to regulate transcription. To understand the molecular basis for such isoform-specific functions we have, for the first tim… Show more

“…Also, RNAi-mediated downregulation of ACTN4 showed an association between the over expression of ACTN4 and an aggressive phenotype of oral squamous cell carcinoma [37]. Recently, the protein was found to be associated with several factors with varying functions, suggesting other roles for this protein, apart from actin regulation [38]. Indeed, Khurana et al identified a LXXLL motif that was responsible for interaction of ACTN4 with the estrogen receptor and co-activators [39].…”

“…3G), which may restrict the reorganization or alignment of actin stress fiber and thus the formation of cell polarity. Recently, ACTN1-ACTN4 heterodimers have been suggested to be the most abundant form in many cell lines and may have different properties and interacting proteins compared with homodimerized ACTN1-ACTN1 and ACTN4-ACTN4 (31). Thus, the unavailability of heterodimerized ACTN1-ACTN4 in ACTN4 KD cells may also limit the function of ACTN1 in the maintenance of melanoma cell morphology.…”

“…Issues to consider as network-based studies of neuropsychiatric disorders move forward Molecular interactions, pathways, and networks are context-specific Whether two molecules interact strongly depends on their biological context, so molecular networks in general, and modules in particular, are context-specific. They can be tissue-specific [23,109], allele-specific [45,93,110], splicing isoform-specific [111], cell-type-specific [43], or brain layerspecific [41,42]. They also vary over time.…”
