Although it has been speculated that seeds of the gymnosperm family Podocarpaceae have an underdeveloped embryo, no detailed studies have been done to definitively answer this question. Our purpose was to determine if embryos in seeds of two species of Podocarpaceae, Podocarpus costalis and Nageia nagi, from Taiwan are underdeveloped and to examine the kind of dormancy the seeds have. Embryos in fresh seeds of P. costalis were 4.6^0.5 mm long, and they increased in length by about 54% before radicle emergence (germination), demonstrating that the embryo is underdeveloped at seed maturity. Seeds germinated to . 90% at 30/20, 25/15 and 258C in light in # 4 weeks, without any cold stratification pretreatment. Thus, seeds of P. costalis have morphological dormancy (MD). Embryos in fresh seeds of N. nagi were 7.4^0.8 mm long and they increased in length by about 39% before radicle emergence (germination) occurred, indicating that the embryo is underdeveloped at seed maturity. Seeds germinated to , 25% at 30/20 and 258C in light in 4 weeks but to . 90% at the same temperatures in 12 weeks. Thus, most seeds of N. nagi have morphophysiological dormancy (MPD). Although underdeveloped embryos are considered to be a primitive condition in seed plants, they also occur in the most advanced orders. The occurrence of underdeveloped embryos in Podocarpaceae documents that they are not restricted to a basal clade in gymnosperms.

The most critical phase in plant life is the germination period, which is influenced by both intrinsic and environmental factors. Assessment of cold stratification on several endogenous hormone, IAA, abscisic acid (ABA), gibberellin GA1/3 (GA1/3), zeatin-riboside (ZRs) and isopen-tenyl adenine (iPAs), and germination of Cornus walteri Wanger. seeds was done. Relationship between endogenous hormone and seed germination and mechanism of seed dormancy of C. walteri were also analysed. The results showed that the significant fluctuatation of both IAA and iPAs content was fond during cold stratification period, while the variation of ZRs was little, that of both ABA and GAs content increased with old stratification days. Effects of cold stratification on both GR and GP were significant (p < 0.05), which play an important role in releaving of seed germination and improving seed germination. The GR and GP were significantly negatively correlated with the contents of ABA and GA1/3, and positively correlated with the following iPAs, ZRs/ABA, iPAs/ABA.

Bamboo is a versatile, arborescent, perennial and non-wood forest tree with tremendous commercial importance. For mass scale propagation of bamboo, the micropropagation is an effective way for producing elite, infection free and true-to-type planting material. Here, the nodal explants of Dendrocalamus strictus used to develop an effective protocol for micropropagation based on tissue culture technique. In this study, the sterilization treatment of 70% ethanol + Tween 20 + Bavistin + Hgcl2 + PPM was successfully controlled the contamination up to 90 % as compared to other treatments. The shoots were initiated from nodal segments in MS medium supplemented with BAP (4 mg/l) and PPM (500µl/l). Shoot multiplication was found best with BAP (4 mg/l) and kinetin (2 mg/l) by using liquid MS medium. Whereas, rooting in solid MS medium has shown good results when supplemented with NAA (4mg/l). Healthy and disease-free plants were obtained after hardening under greenhouse conditions. Genetic fidelity testing by using ISSR markers reported that there was no variation in plantlets developed through micropropagation.
