An Address System in the Vasculature of Normal Tissues and Tumors

Abstract: The vasculature of individual tissues is highly specialized. The endothelium in lymphoid tissues expresses tissue-specific receptors for lymphocyte homing, and recent work utilizing phage homing has revealed an unprecedented degree of specialization in the vasculature of other normal tissues. In vivo screening of libraries of phage that displace random peptide sequences on their surfaces has yielded specific homing peptides for a large number of normal tissues. The tissue-specific endothelial molecules to whic… Show more

“…In vivo proteomic approaches have revealed a striking array of vascular bed-specific phenotypes (reviewed in Rajotte 2000 andSimonson andSchnitzer 2007). For example, antibody and subfractionation strategies have been used to generate monoclonal antibodies that specifically target the caveolae in one vascular bed or another (McIntosh et al 2002).…”

“…A new approach is therefore to develop agents that induce apoptosis in androgen-independent prostate cancer cells, in a proliferation-independent manner. Several of these agents are under preclinical development, and include PSA-activated prodrugs 80,81 and targeted anti-angiogenic agents 82 . Several are also in early clinical trials.…”

“…Alternatively, the angiogenic response of the endothelium to VEGF may be host organdependent. Indeed, endothelial cells have organspecific phenotypes and specialized properties Ruoslahti and Rajotte, 2000). Finally, the metabolic organ microenvironment and its interaction with neoplastic cells may modulate the VEGF response in vivo.…”
