An Accurate Method for Estimating Sizes of Small and Large Plasmids and DNA Fragments by Gel Electrophoresis

Abstract: Several regression methods were tested for estimating the sizes of a wide range of plasmids (1.37-312 MDa) and restriction fragments (2.2-14.2 MDa) by agarose gel electrophoresis. The most accurate and least variable method was the multiple regression of log,, molecular size against log,, relative mobility and the reciprocal square root of the relative mobility. This method gave a good fit to all the data with low percentage errors of the molecular size estimates ( I 3.0 & 1.5 %). It is suggested that with thi… Show more

“…Gels were stained in 2 mg/ml ethidium bromide (Sigma, Brondb, Denmark), illuminated with ultraviolet light at 254 nm using an UV transilluminator (UVP, model TS-40, San Gabriel, CA, USA) and photographed with a Polaroid Land Camera (Polaroid, MP-4, SigmaAldrig, Broenby, Denmark). Plasmid mobilities were calculated relative to the reference plasmids in E. coli strains V517 (Macrina, 1978) and 39R861 (Threlfall et al, 1986), and sizes were estimated in accordance with the methods of Rochelle et al (1986). Coiled covalently closed and open circular DNA molecules were differentiated according to Hintermann et al (1981).…”

“…Visualization was made by staining with ethidium bromide (Sigma, 2 pg ml-' distilled water or 3 pg ml-' gel) followed by exposure to UV-light (254 nm) (Olsen et al 1990). Approximate molecular size of the plasmids was estimated from the migration patterns of supercoiled plasmids in 2 reference strains of Escherichia coli, V517 (Macrina et al 1978) and 39R861 (Threlfall et al 1986), using the regression equation recommended by Rochelle et al (1985).…”
