Amygdalin Analogues Inhibit IFN-γ Signalling and Reduce the Inflammatory Response in Human Epidermal Keratinocytes

Abstract: Peptide T (PT), an octapeptide fragment located in the V2 region of the HIV-1 gp120-coating protein, appears to be beneficial in the treatment of psoriasis. Our previous investigations suggest that keratinocytes play a key role in conditioning the therapeutic effects of PT in psoriasis. The aim of this study was to explore the effects of PT and the peptidomimetic natural products, Dhurrin and Prunasin, on the expression of the IL-6, IL-8, IL-23, HSP70 and ICAM-1 on IFN-γ and TNF-α-NHEK activated cells. “…Recent studies [23] have reported that AG markedly improved glucose tolerance, and ameliorates insulin action in obese and/or diabetic patients by activating a signalling pathway related to glucose metabolism. In addition, a large body of evidence demonstrates that AG plays an important role in enhancing insulin sensitivity and reducing hyperlipidaemia [24]. Therefore, it is reasonable to speculate that this mechanism has the potential to treat acute liver injury in patients.…”

“…The prunasin analogues, amygdalin and manderonitrile, were also quantified in plasma at high concentrations. Prunasin has been demonstrated to inhibit the production of pro-inflammatory cytokines by human keratinocytes [ 28 ], implying a possible anti-inflammatory action. The following compounds were also identified in plasma at relatively-high concentrations; tumulosic acid, paeoniflorin, 4- O -methylgallic acid, gallic acid, manderonitrile, and ( E )-cinnamic acid.…”

“…Randomized controlled trials of XFZY have demonstrated valid effects in patients with TBI 36 37 38 . XFZY - derived compounds, including Hydroxysafflor yellow A (HSYA) and Amygdalin, have been shown to possess an ability to control the inflammatory response 39 40 41 . The above evidence also indicates that the potential mechanism of neuroprotection against TBI by XFZY could involve the reduction of the inflammatory cascade.…”
