Amplified Immunoassay ELISA-ELCA for Measuring Clostridium botulinum Type E Neurotoxin in Fish Fillets

Abstract: The measurement of Clostridium botulinum type E toxin in fish was accomplished using an amplified immunoassay (enzyme-linked immunosorbent assay-enzyme-linked coagulation assay [ELISA-ELCA]) based on the coagulation cascade. Fresh catfish fillets inoculated with a mixture of spores from five strains of C. botulinum type E were packaged in high barrier film with air, vacuum and modified atmosphere and stored at 4, 8 or 16°C for up to 75 days. Toxin production was monitored during storage by both mouse bioassay … Show more

“…The sensitivity of the conventional ELISAs for botulinum neurotoxin detection is some 10-to 100-fold lower than that of the mouse bioassay (47,164,174,177) (Table 2). Signal amplification using a chromogenic diaphorase system (189), biotinylated antibodies and avidin-enzyme conjugate (66,68), or enzyme-linked coagulation assay (ELCA) (53,54,178) has enabled increased assay sensitivity.…”

“…The clear filtrate was tested for the presence of C. botulinum toxin by standard mouse bioassay (FDA, 1992) with minor modifications to reduce trypsin solution concentration (5% trypsin solution used instead of 10%). It was also tested by amplified enzymelinked immunosorbent assay -enzyme-linked coagulation assay (ELISA-ELCA) (Roman et al, 1994). The filtrate was divided into three aliquots.…”
