Amplified Detection of Transcriptional and Translational Inhibitors in Bioluminescent Escherichia coli K-12

Abstract: The excessive prescription of antimicrobial agents and their use as animal growth promoters lead to the spread of resistance among pathogenic bacteria. Consequently, unnecessary use should be minimized, and new chemicals with novel mechanisms of action are needed. The authors have developed a fast method to measure the activity of antibiotics by means of a genetically engineered strain of Escherichia coli K-12. The system is based on the full-length bacterial luciferase operon coupled to the tetracycline-induc… Show more

“…Differences in reporter gene expression versus the numbers of CFU have been noted previously, particularly with specific classes of drugs, such as fluoroquinolones and ␤-lactams (19,27). The sensitivity of LORA to fluoroquinolones was enhanced by extending the anaerobic incubation time from 7 to 10 days (data not shown), but while this decreased the MIC obtained by LORA, a discrepancy between luciferase assayand CFU-based MICs remained.…”

“…Such systems were created on the basis of beta-galactosidase (lacZ) (5,36), green fluorescent protein (GFP) (18,26,29), and luciferase operon (lux) (1,13,19,26,27,35,41) reporter genes. For the comparative studies using reporter constructs, especially under conditions of global repression of gene expression, it is highly beneficial to use an additional reporter gene as a reference.…”

“…In these "reverse" approaches, luciferase expression provides a control signal which is inhibited when the cells are incubated with transcriptional/translational inhibitors [37][38][39].
