AMPK: energy sensor and survival mechanism in the ischemic heart

Abstract: AMP-activated protein kinase (AMPK) is a critical regulator of cellular metabolism and plays an important role in diabetes, cancer and vascular disease. In the heart, AMPK activation is an essential component of the adaptive response to cardiomyocyte stress that occurs during myocardial ischemia. During ischemia-reperfusion, AMPK activation modulates glucose and fatty acid metabolism, mitochondrial function, endoplasmic reticulum stress, autophagy and apoptosis. Pharmacological activation of AMPK prevents myoc… Show more

“…Recent studies showed that restoration of AMPK activation reduced oxidative stress [18, 57], while other studies have linked AMPK stimulation to enhanced oxidative stress [61, 62]. Although the exact relationship between AMPK and oxidative stress is a matter of debate, our results support the notion that restoration of AMPK activation contributes to the amelioration of oxidative stress injury.…”

“…AMPK is an “endogenous survival mechanism”[57], and its activation plays a vital role in protecting against diabetes mellitus [58] and myocardial IR injury [14]. AMPK has been reported to exert an infarct-sparing effect by increasing glucose uptake and glucose transporter-4 translocation [14] and precluding cardiomyocyte apoptosis [14, 59].…”

“…Brian Black (University of California, San Francisco, USA) discussed transcriptional regulation of Prkaa2, which encodes the alpha2 subunit of AMP-activated protein kinase (AMPK), a key therapeutic target in metabolic syndrome, type 2 diabetes and cardioprotection (Qi and Young, 2015). Epigenetic datasets were mined for evolutionary conservation of non-coding sequences within 100 kb of the Prkaa2 start site, and a predicted cardiac enhancer was identified.…”

“…23,24 AMPK signaling is required for angiogenesis in vivo and in vitro. [25][26][27][28] However, there is no information on the consequences of AMPK deletion in arteriogenesis. In this study, we sought to examine the role © 2016 American Heart Association, Inc. Objective-AMP-activated protein kinase (AMPK), an energy and redox sensor, is activated in response to various cellular stresses, including hypoxia, nutrient deprivation, oxidative stress, and fluid shear stress at the site of vessel blockade.…”
