Amoebicidal activity of curcumin on Entamoeba histolytica trophozoites

Abstract: This is the first report evaluating the effectiveness of curcumin against E. histolytica. Our results suggest that CUR could be considered for evaluation in future pharmacological studies as a promising amoebicidal agent or as complementary therapy.

“…CUR, the active component of turmeric, has received considerable interest as a potential therapeutic agent for the prevention or treatment of various diseases, it possesses a wide range of pharmacological properties, for example anticancer [ 30,31 ] and antiparasitic activity. A recent work shows that CUR reduces growth in E. histolytica [ 10 ] and in G. lamblia. [ 11,32 ] However, although CUR shows excellent pharmacological qualities, its clinical use is limited, due to the low solubility in water [ 12,13 ] ; thus, limiting their bioavailability and poor systemic absorption in the digestive tract, this promotes poor pharmacokinetics in vivo , which affects their pharmacological effect.…”

“…Several works have shown that CUR exhibits also anticancer activity, in Caco‐2 cells CUR affects the growth and cell viability. [ 8,9 ] Recently, Rangel‐Castañeda and coworkers demonstrated that 100–300 µ m of CUR has amoebicidal activity, affecting Entamoeba histolytica trophozoites, growth, and morphology, in a dose‐dependent behaviour, [ 10 ] in Giardia lamblia CUR inhibits the growth by affecting the microtubules; dramatic changes on cell morphology and loss of attachment were observed. [ 11 ]…”

“…Likewise, curcumin has been shown to possess antiparasitic activity against malaria and Giardia lamblia [32, 33]. Recently, it was determined by trypan blue exclusion test that curcumin affects the growth and cell viability of E. histolytica , in a time- and dose-dependent manner [34]. In the present work, we studied the antiamoebic activity of curcumin in the ALA model in hamster, and our results suggest that curcumin could help to greatly decrease the incidence of this type of infection.…”

“…Dye exclusion assay was used to determine viability of trophozoites as previously described. 43,44 Briefly, after incubating neutrophils with E. histolytica trophozoites for 3 h as described in the NET formation assay, cells were collected in a 1.5 ml Eppendorf tube and centrifuged at 5000 rpm in a microcentrifuge. Cell pellet was resuspended in 40 l PBS and then 10 l of 0.4% trypan blue (Gibco R ) was added.…”
