Amniotic fluid a crucial role in ensuring the well-being of the fetus during pregnancy and is contained within the amnion cavity, which is surrounded by a membrane. Several studies have shown that volume of amniotic fluid can vary throughout pregnancy and is closely linked to the health and safety of the fetus. This indicates that it is essential to perform accurate measurement and identification of its volume. Obstetric specialist often use a manual method to identify amniotic fluid by visually determining the longest straight vertical line between the upper and lower boundaries. Therefore, this study aims to develop detection model, known as modified Single Deepest Vertical Detection (SDVD) algorithm to automatically measure the longest vertical line by following medical rules and regulations. SDVD algorithm was designed to measure the depth of amniotic fluid vertically by searching the column of pixels that comprised the image sample, excluding any intersection with the fetal body. Performance testing was carried out using 130 images by comparing the manual measurement results obtained by obstetric specialists and the proposed model. Based on the experimental results using modified SDVD, the average accuracy, precision, and recall achieved for amniotic fluid classification were 92.63%, 85.23%, and 95.6%, respectively.

Pixel Classification Based on Local Gray Level Rectangle Window Sampling for Amniotic Fluid Segmentation

This study analyses the use of a pixel classification model to segment amniotic fluid areas on ultrasound (US) images characterized by noise, blurry edge, artifacts, and low contrast. In contrast with the previous methods, this study constrains a training set of pixels based on neighbourhood information with the rectangle window sampling method used to determine the characteristics of each pixel in its environment specifically. The feature extraction is no longer based on the global characteristics of the object rather by taking the value of each pixel in the object area using the sampling window. This research also combines the local first-order statistical methods and gray level information in the window to obtain the characteristics of each pixel. Furthermore, Random Forest and Decision Tree (C.45) were used to classify each pixel into four classes, namely amniotic fluid, placenta, uterus, and fetal body. The classification performance testing of pixel sampling data showed that the Random forest with 5 × 7 window sizes achieved the highest performance at 99.5% accuracy, precision, and recall, respectively. Furthermore, the proposed model was evaluated using 50 new test US images to segment the amniotic fluid area. According to experimental result, proposed models can produce better segmentation area with an increase in the IoU value by 18.3% or a Jaccard coefficient value rate of 0.183 in the range of 0-1 with the previous state of the art method. Furthermore, the proposed model reduces the error rate and improves accuracy by 6.61% and 84.77%, respectively.

The maintenance and self-renewal of hematopoietic stem cells (HSC) in culture is a central focus of hematopoietic stem cell research. In vivo, the balance between HSC differentiation, apoptosis, and self-renewal is regulated at the endosteal surface niche in the bone marrow (BM). In feeder-free cultures, the fate of HSC is affected by growth factors/interleukins and serum, which affect the balance between self-renewal, differentiation, and apoptosis and lead to the rapid loss of multipotent HSC. We report that substituting human amniotic fluid (AF) for serum in HSC cultures provides a growth milieu in which HSC differentiation and apoptosis are down-regulated and multipotent HSC are maintained. Murine BM cells were cultured in serum-free medium containing 25% amniotic fluid and stem cell factor (SCF) only, "AF/SCF" cultures. Compared with serum and multiple growth factor-containing medium, cells cultured for 4 weeks in AF/SCF medium displayed downregulation of differentiation markers while maintaining a high fraction of cells expressing Sca1 (51.8%) and c-kit (10.2%). Reconstitution of lethally irradiated C57BL/6 (Ly5.2) mice with cultured Ly5.1 BM cells resulted in high levels of (cultured) donor cells in primary (78 +/- 19.4% and 94.32 +/- 2.5%, 10(5) and 10(6) cells injected, respectively) and secondary (96.5%) recipients at 8 and 11 months post-transplantation. Hence, long-term repopulation with AF/SCF cultured BM cells was maintained. Addition to the cultures of 10% serum, interleukin (IL)-3, IL-6, granulocyte colony stimulating factor (G-CSF), or granulocyte-macrophage colony stimulating factor (GM-CSF), singly or in combination, resulted in rapid differentiation and apoptosis, leading to the total loss of HSC.
