Aminoacyl‐tRNA synthetase – a molecular multitasker

Swadha Gupta,
Jaykumar Jani,
Vijayasurya
et al.

Abstract: Aminoacyl‐tRNA synthetases (AaRSs) are valuable “housekeeping” enzymes that ensure the accurate transmission of genetic information in living cells, where they aminoacylated tRNA molecules with their cognate amino acid and provide substrates for protein biosynthesis. In addition to their translational or canonical function, they contribute to nontranslational/moonlighting functions, which are mediated by the presence of other domains on the proteins. This was supported by several reports which claim that AaRS … Show more

“…As we all know, amino acids must be activated to obtain extra energy before being incorporated into the peptide chain, and the activation reaction is proceeded under the catalysis of aminoacyl‐tRNA synthetase. [ 33 ] The activated amino acids and tRNA form aminoacyl‐tRNA ensuring the accurate translation of the genetic code and plays an important role in protein synthesis. [ 34 ] Aminoacyl‐tRNA is an important part of the cell wall that can give it strength and shape.…”

“…Tryptophan tRNA synthetase (WARS), catalyzes the addition of the tryptophan amino acid to the cognate tRNA [102]. There are two WARS paralogues that function in the cytoplasm (WARS1) and mitochondria (WARS2), respectively.…”

“…These newly added components broadened aaRS functionality by enabling better recognition of cognate tRNA molecules [10] and proofreading of mischarged tRNAs [11]. Beyond translation, these additional domains contributed to the non-canonical functions of aaRS in gene regulation, cell signaling, inflammation, angiogenesis, tumorigenesis, immune processes, and RNA viral infections [12][13][14][15][16][17][18].…”
