Mycobacterium tuberculosis survival in cells requires mycobactin siderophores. Recently, the search for lipid antigens presented by the CD1a antigen-presenting protein led to the discovery of a mycobactin-like compound, dideoxymycobactin (DDM). Here we synthesize DDMs using solution phase and solid phase peptide synthesis chemistry. Comparison of synthetic standards to natural mycobacterial mycobactins by nuclear magnetic resonance and mass spectrometry allowed identification of an unexpected ␣-methyl serine unit in natural DDM. This finding further distinguishes these pre-siderophores as foreign compounds distinct from conventional peptides, and we provide evidence that this chemical variation influences the T cell response. One synthetic DDM recapitulated natural structures and potently stimulated T cells, making it suitable for patient studies of CD1a in infectious disease. DDM analogs differing in the stereochemistry of their butyrate or oxazoline moieties were not recognized by human T cells. Therefore, we conclude that T cells show precise specificity for both arms of the peptide, which are predicted to lie at the CD1a-T cell receptor interface.

Mahi-mahi Coryphaena hippurus is a promising species for aquaculture development and has been used as a model species for oil toxicology and physiology studies. This species has one of the fastest growth rates of any marine teleost and a unique reproductive biology due to its high spawning frequency and reproductive energy allocation. These characteristics lend the species to being an excellent model for understanding broodstock nutrition for other high energetic pelagic species. In this study, egg morphometrics and larval survival were tracked over a 10-week period from the initial capture of wild mahi-mahi broodstock. Larval quality from subsequent spawns collected over time was quantified using larval survival activity indices (SAIs) as a metric to assess egg quality. Larval SAIs were maintained and did not significantly decrease (p < .05) over the time course of this study. A multiple linear regression based on the elapsed time in captivity of the broodstock, egg diameter and larval SAI at 1 dph provided the most accurate prediction of larval SAI at 3 dph (R 2 = 0.996 p < .05). There were strong positive correlations with larval SAIs at 1 and 3 dph and the key nutrients: eicosapentaenoic acid (C20: 5n-3, EPA) and related fatty acid ratios, vitamin E and nearly all amino acids under investigation. This study demonstrated that larval survival was maintained over time due to the supply of these key nutrients in the broodstock diet. HepatoDyn: A Dynamic Model of Hepatocyte Metabolism That Integrates 13C Isotopomer Data

The liver performs many essential metabolic functions, which can be studied using computational models of hepatocytes. Here we present HepatoDyn, a highly detailed dynamic model of hepatocyte metabolism. HepatoDyn includes a large metabolic network, highly detailed kinetic laws, and is capable of dynamically simulating the redox and energy metabolism of hepatocytes. Furthermore, the model was coupled to the module for isotopic label propagation of the software package IsoDyn, allowing HepatoDyn to integrate data derived from 13C based experiments. As an example of dynamical simulations applied to hepatocytes, we studied the effects of high fructose concentrations on hepatocyte metabolism by integrating data from experiments in which rat hepatocytes were incubated with 20 mM glucose supplemented with either 3 mM or 20 mM fructose. These experiments showed that glycogen accumulation was significantly lower in hepatocytes incubated with medium supplemented with 20 mM fructose than in hepatocytes incubated with medium supplemented with 3 mM fructose. Through the integration of extracellular fluxes and 13C enrichment measurements, HepatoDyn predicted that this phenomenon can be attributed to a depletion of cytosolic ATP and phosphate induced by high fructose concentrations in the medium.
