Background: Vortioxetine is a novel multimodal antidepressant for the treatment of major depressive disorders and is widely used in clinical practice. Vortioxetine has a safe profile. However, there are case reports of other adverse effects in the literature. In this article, a case of amenorrhea due to vortioxetine is presented. Case:The patient is 36 years old, married, female, and is an anesthesiologist. She applied to the psychiatric outpatient clinic with symptoms of major depression. Vortioxetine was initiated at 10 mg/d and then gradually increased to 20 mg/d. She had regular menstruation until now. However, she did not menstruate after taking vortioxetine. The patient used vortioxetine for 3 months and never had a period. Pregnancy test and other tests to investigate organic etiology were normal. Two weeks after discontinuation of vortioxetine, the patient had a menstrual period.Conclusions: Further research is needed on the relationship and possible mechanisms between vortioxetine and amenorrhea.

Duloxetine Amenorrhoea and Hyperprolactinaemia: 2 case reports Two woman developed amenorrhoea and hyperprolactinaemia during treatment with duloxetine. A 41-year-old woman was treated with duloxetine 30 mg/day for major depressive disorder. The dosage of duloxetine was increased to 60 mg/day at the end of first month. In the second month she developed amenorrhoea and her prolactin level was 45.6 ng/mL, which was thought to be related to use of duloxetine. The duloxetine dose was reduced to 30 mg/day and her prolactin level after 1 week went down to 23 ng/mL. Menstruation was detected 10 days later. She continued the treatment with duloxetine 30 mg/day for 5 months. The complaints of amenorrhoea did not reccur, and prolactin levels remained normal.A 47-year-old woman was treated with duloxetine 30 mg/day for major depressive disorder. At the end of the first month she had delayed menstruation and her prolactin level was 57.6 ng/mL. One month later her prolactin level came down to 26.8 ng/mL after treatment with cabergoline. Fifteen days later menstrual period was detected. After normalisation of prolactin, cabergoline was discontinued. Duloxetine treatment was stopped, and she was treated with venlafaxine. The complaints of amenorrhoea did not recur, and prolactin levels remained normal.Author comment: "In our cases, we think that hyperprolactinemia and amenorrhea are related to duloxetine due to the emergence of hyperprolactinemia and amenorrhea after the treatment with duloxetine".Cam B, et al.
