Ameloblastin modulates osteoclastogenesis through the integrin/ERK pathway

Abstract: Proteins of the extracellular matrix often have multiple functions to facilitate complex tasks ranging from signaling to structural support. Here we have focused on the function of one of the matrix proteins expressed in bones and teeth, the matrix adhesion protein ameloblastin (AMBN). Transgenic mice with 5-fold elevated AMBN levels in mandibles suffered from root cementum resorption, delamination, and reduced alveolar bone thickness. AMBN gain of function also resulted in a significant reduction in trabecula… Show more

“…To prepare undecalcified ground sections, tissues were fixed in 4% paraformaldehyde, dehydrated in a graded series of ethanol, embedded in Technovit 7200 (Exakt, Oklahoma, OK, USA) and prepared into 10-mm ground sections for subsequent von Kossa staining. (26) To prepare decalcified paraffin sections, samples were demineralized in 4% EDTA. After paraffin embedding, sections were subjected to H&E staining or immunohistochemistry.…”

“…(26,27) To determine whether cytoskeletal dynamics were involved in the mechanism by which AMBN regulates gene expression, AMBN BMSCs were cultured on BSA-or AMBN-coated culture dishes for 24 hours. Cytoskeletal organization of cultured cells was visualized by F-actin staining using rhodamine-phalloidin (Fig.…”
