Ameliorative Effects of Caffeic Acid Phenethyl Ester on an Eccentric Exercise-Induced Skeletal Muscle Injury by Down-Regulating NF-kB Mediated Inflammation

Abstract: Background and Purpose: Caffeic acid phenethyl ester (CAPE), a phenolic compound isolated from propolis, displays a variety of biological activities. The aim is to examine the protective effect and mechanisms of CAPE on an eccentric exercise-induced muscle injury model. Experimental Approach: An intermittent downhill eccentric exercise protocol was used. The oxidative tissue injury and expression of cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), interleukin-1ß (IL-1ß), monocyte chemotactic p… Show more

“…In the present study, whereas 8-OhDG was strongly expressed in MTX group, reduction in 8-OhDG expression in the group for which MTX was administered together with hesperidin suggested that DNA damage was diminished and, hence, hesperidin had a protective property. NfKB, however, is a molecule which is activated when oxidative stress is developed [45,46] . It was reported that level of NfKB increased with testicular intoxication [47,48] .…”

“…CAPE, which blocked the NF-κB-dependent activation of the inflammatory response (34). In another in vitro study, CAPE significantly suppressed the levels of lipopolysaccharide (LPS)-induced IL-1β, TNF-α and MCP-1 from a macrophage cell line, RAW264.7 (35).…”

“…As a consequence of this inhibition, the activity of COX-1 and -2 and the activation of the COX-2 gene expression are suppressed (32,33). Recently, the protective effect of CAPE on a model of eccentric exercise-induced muscle injury was investigated (34). The study results showed that inflammatory skeletal muscle injury enhanced the expression of COX-2 and iNOS, as well as the production of IL-1β and monocyte chemoattractant protein-1 (MCP-1).…”

“…EXERCISE INDUCES INFLAMMATORY and stress signaling in muscle, including NF-B and JNK activation (17,39). These pathways drive the expression of proinflammatory genes such as IL-6 and chemokines such as chemokine C-C motif ligand 2 (CCL2)/monocyte chemoattractant protein-1 (MCP-1), and chemokine C-X-C motif ligands 1 and 5 (CXCL1/5) (23,33,39,43,44).…”
