Amelioration of Radiation Enteropathy by Dietary Supplementation With Reduced Coenzyme Q10

Abstract: Purpose Effective methods to ameliorate radiation enteropathy have not been developed. To address this issue, we investigated the reduced form of coenzyme Q10 (rCoQ10) as a potential radioprotector in a mouse model. Methods and Materials rCoQ10 was added to a standard laboratory mouse diet at a final concentration of 1.0% 9 days before irradiation and 30 days thereafter or dissolved in corn oil and administered transorally. Accumulated amounts of coenzyme Q10 (CoQ10) or… Show more

“…Cells were exposed to 5 or 8 Gy 150 kV X-rays delivered at 0.57 Gy/min using an MBR-1505R2 generator (Hitachi Ltd.). The beam was filtered through a 1-mm aluminum board and the accuracy of irradiation was checked, as previously described ( 30 ).…”

“…X-ray irradiation was performed using the MBR-1505R2 X-ray generator (Hitachi, Tokyo, Japan) at a voltage of 150 kV and a current of 5 mA with a 1-mm thick aluminum filter (0.5 Gy min −1 at the target). Prior to each experiment, the mice were anesthetized using an intraperitoneal administration of somnopentyl (0.1 mg g −1 body weight), and then immobilized in a customized harness that exposed the tumor while shielding the remainder of the body with lead during irradiation, as described previously [ 31 ].…”

“…The anesthetic Somnopentyl (0.1 mg/g body weight) was intraperitoneally administrated prior to radiation exposure. A lead shield covering the head and chest of each mouse was used prior to x-ray irradiation [ 21 ].…”

“…Dihydroethidium (DHE) was measured in the intestinal tissue as previously described [ 21 ]. In brief, DHE was dissolved in dimethyl sulfoxide and diluted with PBS immediately before use.…”
