Ameliorating effects of agomelatine on testicular and epididymal damage induced by methotrexate in rats

Abstract: The aim of this study was to investigate the possible prophylactic effects of agomelatine (AGO) against testicular and epididymal damage induced by methotrexate (MTX) in rats. Twenty‐four male Wistar albino rats were divided into three groups: Group I (control group), Group II (MTX group: 20 mg/kg MTX, i.p, single dose), and Group III (MTX+AGO group: 20 mg/kg MTX, i.p, single dose+40 mg/kg AGO; gavage, 7 days). The rats were killed under anesthesia 24 hours after the last AGO application. Testicular and epidid… Show more

“…Testicular injury is a serious adverse effect of MTX therapy, thus the preservation of germinal cells is an essential target during cancer chemotherapy [ 6 , 7 ]. In line with previous reports [ 22 , 23 ], MTX treatment in the current study was associated with a significant decrease in testicular weight, indicative of testicular atrophy.…”

“…Oxidative stress, which arises as a result of the imbalance between reactive oxygen species (ROS) and the endogenous antioxidant system, is an important factor in MTX-induced testicular injury [ 6 , 7 , 22 , 25 ]. MTX can increase ROS production by the elevation of homocysteine [ 28 ], reduction of NADPH [ 29 ], and activation of NADPH oxidase [ 30 ].…”

“…[20] For example, the signs of cardiovascular impairment such as arterial hypotension, arrhythmia, and conduction disorder occur instantly, while symptoms like obesity, anemia, or metabolic disorders associated with glucose can only be detected with an n-week delay. [21,22] The long-term effects are easier to treat compared to the acute ones that emerge immediately after poisoning and progress within the period of 3 to 5 days. [4,23] The prolonged use of antipsychotics, even in doses that do not cause poisoning, results in low blood pressure, due to the effect on the autonomic nervous system.…”

“…Methotrexate (MTX) is a folic acid antagonist used as a medication through the toxic effects of MTX [6][7][8]. It has been reported that male individuals exposed to MTX exhibit oligozoospermia and structural chromosomal rearrangements [9].…”
