Arsenic trioxide (As 2 O 3 ) is an effective therapy in acute promyelocytic leukemia (APL), but its use in other malignancies is limited by the higher concentrations required to induce apoptosis. We have reported that trolox, an analogue of a-tocopherol, increases As 2 O 3 -mediated apoptosis in a variety of APL, myeloma and breast cancer cell lines, while nonmalignant cells may be protected. In the present study, we extended previous results to show that trolox increases As 2 O 3 -mediated apoptosis in the P388 lymphoma cell line in vitro, as evidenced by decrease of mitochondrial membrane potential and release of cytochrome c. We then sought to determine whether this combination can enhance antitumor effects while protecting normal cells in vivo. In BDF 1 mice, trolox treatment decreased As 2 O 3 -induced hepatomegaly, markers of oxidative stress and hepatocellular damage. In P388 tumor-bearing mice, As 2 O 3 treatment prolonged survival, and the addition of trolox provided a further significant increase in lifespan. In addition, the combination of As 2 O 3 and trolox inhibited metastatic spread, and protected the tumor-bearing mice from As 2 O 3 liver toxicity. Our results suggest, for the first time, that trolox might prevent some of the clinical manifestations of As 2 O 3 -related toxicity while increasing its pro-apoptotic capacity and clinical efficacy in hematological malignancies.

Continuous exposure of humans to arsenic through long-term ingestion of contaminated drinking water and its attendant health problems has been widely reported. It is also known that arsenic interact with other substances, metals inclusive thereby potentiating its effects or vice versa. In this study, we examined the effects of sodium arsenite (SA) and lead acetate (LA) in wistar rats. Sodium arsenite (2.5mg/kg bd.wt) and lead acetate (14mg/kg bd.wt) were fed to rats by gavage for fourteen consecutive days alone or simultaneously. Control rats were fed with distilled water. Clastogenic effects were observed in the bone marrow cells using the micronucleus assay. In addition, serum activities of gamma glutamyl transferase (γ-GT), alkaline phosphatase (ALP), alanine amino transferase (ALT ) and aspartate amino transferase (AST) were monitored . The findings indicate that SA and LA separately induced the formation of micronucleated polychromatic erythrocytes (mPCEs) in the bone marrow of the rats significantly (P < 0.05) by about 9 and 8 folds respectively. When fed simultaneously, the induction was about 22 folds as compared with the negative control group. SA significantly induced the serum activity of all the enzymes while LA significantly induce the activity of only γ-GT and ALP (P < 0.05). Simultaneous feeding of SA and LA also markedly induced the activity of all the enzymes in the serum. Mild infiltrative haemorrhage was observed in the lungs of rats exposed to the two compounds. This study underscores the enhanced toxic effect of combined or simultaneous exposure to toxic substances.

Objective: Arsenic, one of the major natural as well as anthropogenic pollutant produces biochemical changes in the organs of animals due to its potential toxicity. Arsenic is toxic to fish and may be taken through gills and skin. In the current work, arsenic trioxide-treated fishes were prescribed with potentized homoeopathic drug, ars alb 30C, and ars alb 200C. The biochemical contents in exposed gill tissues of zebra fish (Danio rerio) were analysed using Fourier transform infrared spectroscopy (FTIR). Methods:The acclimatised fish were treated with arsenic trioxide and LC50 was calculated. Along with arsenic trioxide, the homoeopathic drug ars alb 30C and ars alb 200C were given three drops per litre. After a period of 7 and 14 d, the gills of exposed fish were dissected. The samples were lyophilized and analysed in FTIR.Results: In arsenic exposed fish, the amine N-H stretch was different when compared to the control and the treated different potencies of ars alb 30C and 200C respectively at 7days. Whereas at 14 d the arsenic exposed fish the amine N-H stretch was drastically reduced. Notably, in treating different potencies of ars alb 30C and 200C, the amine N-H stretch was increased. Conclusion:It was found that the higher the potency that is ars alb 200C was more effective against Arsenic toxicity. It shows higher dilution may be efficacious.
