Amantadine in a Multimodal Analgesic Regimen for Alleviation of Refractory Osteoarthritis Pain in Dogs

Abstract: Background: Nonsteroidal anti-inflammatory drugs (NSAIDs) do not always provide sufficient pain relief in dogs with osteoarthritis (OA).Hypothesis: The use of amantadine in addition to NSAID therapy will provide improved pain relief when compared with the use of nonsteroidal analgesics alone in naturally occurring OA in dogs.Animals: Thirty-one client-owned dogs with pelvic limb lameness despite the administration of an NSAID. Methods: The study was randomized, blinded, and placebo controlled with parallel gro… Show more

“…The CSOM is a subjective instrument that satisfied some psychometric qualities in this clinical trial. The owner-described behaviors in the CSOM were similar to previously described behaviors in canine studies of OA symptoms and pain [7,19]. These similarities suggest the face validity k of the CSOM, which indicates that this measurement assesses the appropriate outcome [32].…”

“…The variety of OA signs suggests the need for the implementation of a clinical assessment that includes a broader variable profile than the current standardized OA profile. For this purpose, the client-specific outcome measures (CSOM) require the owner to report pain behavioral changes and/or impaired activities in their OA-afflicted dogs [18,19]. However, the promising CSOM have not met the technical validity b criteria [19].…”

“…In humans, multimodal analgesia has been shown to decrease post-operative morbidity and mortality, improve quality of life and patient satisfaction, and decrease the associated costs to hospitals and insurance companies (Skinner, 2004). In addition to the general agreement of a clinical benefit to this approach (Lascelles & Main, 2002), there are also an increasing number of research studies in non-verbal species supporting multimodal analgesia (Lascelles et al, 2008; Slingsby, Murrell & Taylor, 2010; Martins et al, 2010). One simple way to include multimodal analgesia is the incorporation of a local anesthetic to desensitize a specific region, in combination with systemic analgesic administration.…”

“…Owners then rate the degree of impairment. Use of this method has demonstrated that owners were able to assess differences in their pet's mobility when an analgesic was administered compared to a placebo (50). The use of CSOM has been shown to be a sensitive method for use in the pain management of dogs (51).…”
