ALW peptide ameliorates lupus nephritis in MRL/lpr mice

Abstract: BackgroundLupus nephritis (LN) is a common and serious complication of systemic lupus erythematosus. Anti-double-stranded (ds) DNA immunoglobulin G (IgG) plays a pivotal role in the pathogenesis of LN. Currently, there are various therapies for patients with LN; however, most of them are associated with considerable side effects. We confirmed previously that ALW (ALWPPNLHAWVP), a 12-amino acid peptide, inhibited the binding of polyclonal anti-dsDNA antibodies to mesangial cells and isolated glomeruli in vitro.… Show more

“…This confirms that lupus nephritis initiation is not due to a single factor [ 22 ]. Nonetheless, a large amount of research suggests that anti-dsDNA antibodies accelerate lupus nephritis processes, and lupus nephritis symptoms are improved by blocking anti-dsDNA antibodies [ 23 , 24 ]. We conclude that anti-dsDNA antibodies are involved in renal inflammation and fibrosis; however, further investigation is needed on the complex mechanism of the cytokine networks and signaling pathways.…”

“…This molecule can also prevent IgG isotypes or anti-dsDNA antibodies in SLE patients’ serum interacting with glomerulus and glomerular mesangial cells [ 24 ]. ALW peptides can inhibit glomerular deposition of antibodies, reduce serum anti-dsDNA antibody titers, improve renal pathological manifestations, or suppress proliferation, fibrosis, and inflammatory cell infiltration of renal tissue, providing a protective effect on lupus nephritis manifestation in MRL/lpr mice [ 23 ].…”

“…Furthermore, deposition of immune reactants plays a pivotal role in the occurrence, development, and prognosis of LN as well (30). The cross-reaction between immunoglobulin (IgG/IgM) and renal antigens plays key regulatory roles in many biological processes, such as activation of complement cascades, regulation of gene expression and cell proliferation (31). And it induces phenotypic changes of glomerular resident cells, leading to kidney injury in LN (32).…”

“…Administration of nucleosomal peptides to SNF1 mice delayed the onset of nephritis and improved survival (140); in the mechanistic analysis, an increment of regulatory T cells was shown, and a direct effect on B cells was not investigated. Finally, peptides that bind anti-DNA antibodies can prevent their pathogenicity in vivo (141,142).…”
