Alternative Neural Crest Cell Fates Are Instructively Promoted by TGFβ Superfamily Members

Abstract: How growth factors influence the fate of multipotent progenitor cells is not well understood. Most hematopoietic growth factors act selectively as survival factors, rather than instructively as lineage determination signals. In the neural crest, neuregulin instructively promotes gliogenesis, but how alternative fates are determined is unclear. We demonstrate that bone morphogenic protein 2 (BMP2) induces the basic-helix-loop-helix protein MASH1 and neurogenesis in neural crest stem cells. In vivo, MASH1+ cells… Show more

“…Although TGFb1 induces a smooth muscle cell fate in NCSCs on prolonged incubation [21], smooth muscle actin (SMA)-expressing smooth muscle cells were not present after 4 hours of TGFb1 priming (data not shown). Furthermore, short-term TGFb1 treatment did not affect cell survival (data not shown).…”

“…29 Cultured neural crest cells differentiate towards spindle-shaped SMCs when treated with TGF-β. 30 Moreover, TGF-β1 signalling has recently been shown to be required for SMCspecific gene expression in embryoid bodies. 31 In contrast to PDGF, TGF-β isoforms clearly promote the contractile phenotype in adult SMCs.…”

“…Wnt signaling on its own instructively promotes sensory neurogenesis in eNCSCs [19,20]. BMP signaling alone, on the other hand, instructively drives eNCSCs into the autonomic lineage in vitro [21], while in vivo BMP signaling regulates proliferation, differentiation and survival of NC-derived cells in a lineage specific fashion [22][23][24]. Cells with NCSC-like features can also be found in various postmigratory target structures of the neural crest, like in the embryonic sciatic nerve [25], adult skin [26,27], gut [28], adult dorsal root ganglia (DRG), whisker pad, and bone marrow [29].…”
