Altered Response of Intestinal Mucosal Fibroblasts to Profibrogenic Cytokines in Inflammatory Bowel Disease

Abstract: These results suggest the presence of an activated subpopulation of fibroblasts in both UC and CD tissue irrespective of the presence of tissue fibrosis or disease type.

“…These CD fibroblasts may be stimulated by local IGF-1 to produce collagens. In inflammatory bowel disease, fibroblasts isolated from affected intestinal mucosa display faster doubling time and greater proliferation in response to mediators (including IGF-1) and secrete higher levels of collagen compared to control fibroblasts [31]. As well as this increased expression of IGF-1R in fibroblast-like cells in CD, our data also showed that numerous IGF-1R positive IC are found at margins of fibrosed muscularis propria and within septas.…”

“…Interestingly, the pro-apoptotic effect of pro-inflammatory cytokines was only observed with myofibroblasts isolated from normal mucosa, myofibroblasts isolated from inflamed www.intechopen.com mucosa being apoptosis-resistant (Francoeur et al 2009). Distinct intrinsic properties of myofibroblasts isolated from inflammatory bowel disease vs non-inflammatory bowel disease patients remains to be elucidated but is not without precedent (Lawrance et al 2001). The contribution of bone marrow-derived myofibroblasts in the regenerative process in inflammatory bowel disease (Andoh et al 2005, Brittan et al 2007) may explain the resistance to cytokine-mediated apoptosis.…”
