Altered cytoskeletal structures in transformed cells exhibiting obviously metastatic capabilities

Abstract: Cytoskeletal changes in transformed cells (LM-51) exhibiting obviously metastatic capabilities were investigated by utilization of double-fluorescent labelling through combinations of: (1) tubulin indirect immunofluoreseonce plus Rhodamine-phalloidin staining of F-actins; (2) indirect immunofluorescent staining with α-actinin polyclonal-and vinculin monoclonal antibodies. The LM-51 cells which showed metastatic index of >50% were derived from lung metastasis in nude mice after subcutaneous inoculation of human… Show more

“…In comparison with healthy cells, biomechanical investigations reported some common features of many types of tumor cell lines, such as a less structured CSK [7][8][9][10] with lower cell mechanical [10][11][12] and cyto-adhesive properties [9,10,13,14]. All factors which, from a biophysical point of view, augment the metastatic potential of the cancer cells [3,7,9,14,15].…”

“…These include immortalization of retroviral transduction by simian virus 40 (SV40) T antigen [ 37 , 38 ], Cdk4R24c/CyclinD1 [ 39 ], and/or human papilloma virus 16 E6/E7 [ 40 ]. Immortalized cells increase the risk of tumor formation, aneuploidy [ 41 ], and structural rearrangements [ 42 ]. Recently, the establishment of untransfected HCEC line [ 43 ] and immortalization of HCECs with human telomerase reverse transcriptase have been explored [ 44 ].…”

“…The cytoskeleton is a dynamic network of filamentous proteins that controls cell shape, maintains intracellular organization and is involved in cell motility ( 1 , 2 ). Alterations in cytoskeletal structures serve a pivotal role in controlling cancer cell behavior, including cell adhesion, migration and invasion ( 3 , 4 ).…”
