Alterations of gene expression of RB pathway in Opisthorchis viverrini infection-induced cholangiocarcinoma

Abstract: Opisthorchiasis has the significant relationship with the high prevalence of cholangiocarcinoma (CCA; a bile duct cancer) in the endemic areas in Southeast Asia. To reveal the molecular mechanism of the tumorigenesis induced by Opisthorchis viverrini infection, the present study investigated the kinetic expression of RB pathway genes, including RB1, p16(INK4), cyclin D1, and CDK4, during the development of opisthorchiasis-associated CCA in hamster model. The results of quantitative real-time polymerase chain r… Show more

“…The evidence from epidemiologic investigations and animal experiments has confirmed the definite relationship between O. viverrini infection and prevalence of CCA, combining with the traditional eating custom of fermented food containing potent carcinogens, for example, N-nitrosodimethylamine (NDMA; Sriamporn et al 2004;Poomphakwaen et al 2009;Mitacek et al 1999b;Boonmars et al 2009b).…”

“…Ccnd1 and Cdk4 are key members in Rb1 signaling pathway which plays important role in tumorigenesis. The altered expression of the two genes has been observed in OV-associated CCA (Boonmars et al 2009b). GADD45 is a family of proteins involved in the G2/M cell cycle checkpoint in response to DNA damage (Zhang et al 2002).…”

“…On the other hand, host immunological factors play also a crucial role in determining the outcome of opisthorchiid infection and in initiation of cholangiocarcinogenesis. In this context, it was shown that O. viverrini infection down-regulates RB1 (retinoblastoma 1) and p16 INK4 (cyclin-dependent kinase inhibitor 2A) expression and up-regulates cyclin D1 and CDK4 (cyclin-dependent kinase 4) expression during cholangiocarcinoma development (Boonmars et al, 2009). These proteins are members of the retinoblastoma protein (RB) pathway, which is strongly involved in cancer development.…”

“…The metacercariae of O. viverrini were collected as the naturally infected cyprinoids fish captured from an endemic area of Khon Kaen, northeast Thailand. The parasite species were confirmed under light microscope (Boonmars et al, 2009) and were minced and digested with pepsin-HCl, then filtrated and washed with normal saline.…”
