Alterations in thigh subcutaneous adipose tissue gene expression in protease inhibitor-based highly active antiretroviral therapy

Abstract: Use of protease inhibitor (PI)-based highly active antiretroviral therapy (HAART) has been associated with altered regional fat distribution, insulin resistance, and dyslipidemias. To assess how PI-based HAART affects adipocyte gene expression in male HIV-1-infected patients, reverse transcription-polymerase chain reaction was used to quantify messenger RNA expression of adipocyte transcription factors and adipocytokines in thigh and abdominal subcutaneous adipose tissue from male (1) HIV-1 seronegative subjec… Show more

“…63 There are no studies that have compared regional leptin mRNA expression in subcutaneous adipose tissue; however, there are indirect suggestions that basal expression does not differ between abdominal and gluteofemoral fat. 64,65 Serum leptin increases in parallel with BMI and, interestingly, is higher in women. [66][67][68][69][70] The sexual dimorphism in circulating leptin levels is apparent already in childhood 71,72 and has been attributed to differences in subcutaneous fat mass and adipocyte cell size.…”

“…RNA was isolated by Trizol (Invitrogen) from treated cell lines quantitative real-time PCR was conducted using the method as described previously (37). Briefly, 2 μg of total RNA per sample were converted to cDNA using the SuperScript First-Strand Synthesis system for reverse transcription-PCR (Invitrogen).…”

“…Relative gene expressions of EGFR (NM_207655), VEGFR (NM_008029), FGFR (NM_008010), platelet-derived growth factor receptor (NM_011058), and IGFR (NM_010513) were determined by quantitative real-time PCR analysis (33). The primers shown on Table 5 were designed using PrimerExpress version 2.0 (Applied Biosystems, Foster City, CA).…”
