Alteration of the C Fms Gene in a Blood Sample From a Thorotrast Individual

Abstract: We analyzed six different tissue DNA samples from a leukemic individual who received an injection of Thorotrast for alterations in proto-oncogene or tumor-suppressor gene structure. Our examination of the DNA indicated an alteration of the c-fms gene in the blood sample from this individual. This locus showed a deletion in which the 3' end of the deleted region maps between exons 11 and 12. In this particular case, the type of leukemia is unknown but myeloid leukemia is a neoplasm associated with individuals i… Show more

“…Many studies have shown that IMPDH activity is elevated in tumors and other actively proliferating tissues (3,4,(7)(8)(9)(10)(11)(12)(13)(14)(15). In some cases the elevated activity seems to stem from induction of the type II isoenzyme (9, 11, 12), although similar up-regulation of both isoforms in activated T-lymphocytes has also been reported (14).…”

“…Indeed, inhibition of IMPDH in cultured cells results in decreased guanine nucleotide levels. Ultimately, this leads to the induction of differentiation in neoplastic cells (8) and the suppression of lymphocyte activation (16).…”

“…The abundance of the enzymatic activity is correlated with the rate of growth of mammalian cells (1)(2)(3). Antagonists of this family of enzymes, one of which is mycophenolic acid, serve as medically important antitumor, antimicrobial, and immunosuppressive agents (4).…”

“…Characterization of two IMP dehydrogenase genes in mammalian cells has included an analysis of their promoters and the solution of a crystal structure of rodent IMP dehydrogenase in a complex with mycophenolic acid (5)(6)(7). Mammalian IMP dehydrogenase (IMPDH) 1 II is inducible in a post-transcriptional manner, in response to depressed guanine nucleotide pools (3,8). Saccharomyces cerevisiae has a family of four closely related genes, IMD1, IMD2 (formerly known as PUR5), IMD3, and IMD4, that encode potential IMP dehydrogenases.…”

“…As a negative control, the assay was run in the absence of a precipitating antibody, which resulted in no amplification products for all three of the queried genes (lane 4 of each gel). In addition, PCR for the promoter of type II IMP dehydrogenase, a gene that has not been shown to be responsive to TGF-b or BMP stimulation, yet is critical for mycophenolic acid induced-HL-60 cell differentiation (Collart and Huberman, 1990), resulted in no amplification from the ChIP-recovered DNA (data not shown). Analysis of PMA-induced OPN, Hex and Id2 gene expression by rtPCR (lower panel) revealed that expression patterns are similar to the patterns of Smad6 binding to their respective promoters.…”
