Alteration of respiration capacity and transcript accumulation level of alternative oxidase genes in necrosis lines of common wheat

Abstract: Mitochondrial alternative oxidase (AOX) is the terminal oxidase responsible for cyanide-insensitive and salicylhydroxamic acid-sensitive respiration in plants. AOX is a key enzyme of the alternative respiration pathway. To study the effects of necrotic cell death on the mitochondrial function, production of reactive oxygen species (ROS), respiration capacities and accumulation patterns of mitochondriatargeted protein-encoding gene transcripts were compared between wild-type, lesion-mimic mutant and hybrid necr… Show more

“…Distribution of the Ne1 and Ne2 dominant alleles is postulated to be due to selection advantages such as rust fungus resistance , although no direct evidence proving a relationship between type I necrosis and disease resistance has been reported. In leaves of wheat showing type I necrosis, superoxide is highly accumulated and antioxidant enzymes show high levels of activity (Dalal and Khanna-Chopra 2001;KhannaChopra et al 1998;Sugie et al 2007).…”

“…Total RNA was extracted from the seedling leaves and treated with DNaseI to remove the contaminated DNA. Reverse transcriptase (RT)-PCR with the gene-specific primer sets resulted in amplification of the specific single fragments as described previously (Sugie et al, 2006(Sugie et al, , 2007. RT-PCR for wheat uncoupling protein 1 (WhUCP1) was conducted with the following primers: 5 0 -TGAAAGTGAGATTGCAAGCA-3 0 and 5 0 -GCCCAT-CATTCTTGATTTCA-3 0 .…”

“…Necrotic cell death induced by the Ne1-Ne2 epistatic interaction is accompanied with generation of reactive oxygen (Sugie et al 2007 ). Because the Ne2 gene seems to be located in a chromosomal region closely linked to the R gene against rust fungus, it has been postulated that type I necrosis is due to autoimmune-like responses triggered by the Ne1 -Ne2 interaction (Bomblies and Weigel 2007 ).…”
