Alpha‐adrenergic receptor gene polymorphisms and cardiovascular reactivity to stress in Black adolescents and young adults

Abstract: Cardiovascular reactivity to stress and α-adrenergic receptor (α-AR) function may contribute to the development of hypertension. As Black Americans have an increased risk of hypertension, we evaluated associations between α1A-AR (Arg492Cys), α2A-AR (−1291C/G), and α2B-AR (Ins/Del301-303) gene variants and cardiovascular reactivity in 500 normotensive Black youth. Heart rate, preejection period, total peripheral resistance, and blood pressure were measured during cold and psychological stress. The Arg492Cys pol… Show more

“…Polymorphisms in genes involved in regulation of the adrenergic system, including α 1a AR gene, have been linked to hypertensive disorders [12,24,25]. Functional effects of α 1a AR SNPs might therefore represent a novel mechanism underlying hypertension and heart disease.…”

“…(30) Moreover, it has been associated with other α 2A -AR-related phenotypes suggesting a gain-of-function action such as schizophrenia, weight gain during antipsychotic medication use, body fat accumulation, and vascular reactivity to stress. (3134) The variant is in the promoter region of the gene, and it may exert its effects by influencing gene expression. (35) In keeping with previous findings related to glucose regulation phenotypes,(3134) we found that the G allele was associated with higher admission glucose in Caucasians (P = 0.046) with a similar trend among African-American subjects.…”

“…To address this point, Kelsey et al evaluated associations between three genetic polymorphisms within the α1-, α2A and α2B-ARs genes (Arg492Cys, C-1291G, and Ins/Del301-303, respectively) and cardiovascular reactivity to stress in healthy, normotensive black adolescents as well as in young adults [62]. The psychological stressors used in this study consisted of mental arithmetic tasks and a video game task.…”
