Alpha 1-Antitrypsin Does Not Inhibit Human Monocyte Caspase-1

Abstract: BackgroundAlpha 1-antitrypsin (A1AT) is a 52 kDa serine protease inhibitor produced largely by hepatocytes but also by mononuclear phagocytes. A1AT chiefly inhibits neutrophil elastase and proteinase-3 but has also been reported to have immune modulatory functions including the ability to inhibit caspases. Its clinical availability for infusion suggests that A1AT therapy might modulate caspase related inflammation. Here we tested the ability of A1AT to modulate caspase-1 function in human mononuclear phagocyte… Show more

“…Exogenous AAT can enter the cytoplasm via lipid rafts ( 33 ) and was suggested to directly inhibit the proteolytic activity of casp-1 ( 34 , 35 ). Although the inhibition of cytoplasmic casp-1 of intact cells by AAT was disputed ( 36 ), we wondered if this mechanism is responsible for the AAT-mediated suppression of IL-1β release. This hypothesis was falsified by two independent experiments.…”

“…However, another study indicates that plasma-derived AAT did not reduce caspase-1 activity in the THP-1 macrophage cell line or in cell free extracts, and had no effect on whole human blood production of IL-1β 44. Because IL-1 stimulates caspase-1 gene expression in human blood monocytes45 and caspase-1 cleavage of the IL-18 precursor is IL-1β dependent,46 a reduction in IL-1β will also reduce the level of caspase-1 itself without directly affecting the enzymatic function of caspase-1.…”

“… 32 , 33 The effect of A1AT on caspase-1, a component of inflammasomes, is controversial as some studies demonstrate the inhibitory effect of A1AT on caspase-1 activity, 34 while others did not find any effects. 35 …”
