Aloe-Emodin Induces Mitochondrial Dysfunction and Pyroptosis by Activation of the Caspase-9/3/Gasdermin E Axis in HeLa Cells

Abstract: Aloe-emodin (1,8-dihydroxy-3-hydroxymethyl-anthraquinone), derived from some Chinese edible medicinal herbs, exerts a potential anticancer activity on various cancer cells, making it a drug candidate for cancer therapy. Yet, the role of aloe-emodin in pyroptosis, a new type of cell death, is uncharacterized. In this study, we explored the molecular mechanisms of aloe-emodin-triggered pyroptosis. Aloe-emodin inhibited proliferation and migration and triggered caspase-dependent cell death of HeLa cells in a dose… Show more

“…SRC had similar binding energy to that of its original ligand. AKT1, TP53, ESR1, SRC, and EGFR had energy <−5 kcal/Mol, demonstrating that these five core targets of AE play an important role in the treatment of HCC( Li et al, 2021 ; Liu et al, 2021 ; Li T. et al, 2022 ). The docking results were visualized through PyMOL ( Figures 6 , 7 ).…”

“…Caspase-3 activity was measured as described previously [ 36 ]. Briefly, equal volumes of cell lysates were mixed and incubated with reaction buffer (1 M sodium citrate, 10 mM dithiothreitol, and 50 mM Tris-HCl, pH 7.4) containing Ac-DEVD-AFC (20 μM final) for 30 min at 37 °C.…”

“…Interestingly, the PI3K/AKT pathway also regulates GSDME‐dependent pyroptosis (Yu, Tan, et al, 2022). It may be that the activation of Caspase‐9 is inhibited by AKT, thereby reducing Caspase‐9/3/GSDME axis‐induced pyroptosis (Li, Shi, et al, 2022; Sangawa et al, 2014).…”
