a b s t r a c t B-phycoerythrin (B-PE) was separated and purified from microalga Porphyridium cruentum using onestep chromatographic method. Phycobiliproteins in P. cruentum was extracted by osmotic shock and initially purified by ultrafiltration. Further purification was carried out with a SOURCE 15Q exchange column and analytical grade B-PE was obtained with a purity ratio (A 545 /A 280 ) of 5.1 and a yield of 68.5%. It showed a double absorption peaks at 545 nm and 565 nm and a shoulder peak at 498 nm, and displayed a fluorescence emission maximum at 580 nm. The analysis by sodium dodecyl sulfate epolyacrylamide gel electrophoresis (SDSePAGE) showed a bulky band between 18 and 20 kDa which could be assigned to subunits a and b and a low intensity band of 27 kDa assigned to g subunit. Our protocol provides attractive alternative to consider for the purification procedure to obtain analytical grade B-PE at commercial level.

The conformational state of biliproteins can be determined by optical properties of the covalently linked chromophores. Recently determined crystal structure of truncated form of α-subunit of cyanobacterial phycoerythrin (αC-PE) from Phormidium tenue provides a new insight into the structure-function relationship of αC-PE. To compare their stabilities, we have measured urea-induced denaturation transitions of the full length αC-PE (FL-αC-PE) and truncated αC-PE (Tr-αC-PE) followed by observing changes in absorbance at 565 nm, fluorescence at 350 and 573 nm, and circular dichroism at 222 nm as a function of [urea], the molar concentration of urea. The transition curve of each protein was analyzed for ΔG(D)(0), the value of Gibbs free energy change on denaturation (ΔG(D)) in the absence of urea; m, the slope (=∂∆G(D)/∂[urea]), and C(m), the midpoint of the denaturation curve, i.e. [urea] at which ΔG(D) = 0. A difference of about 10% in ΔG(D)(0) observed between FL-αC-PE and Tr-αC-PE, suggests that the two proteins are almost equally stable, and the natural deletion of 31 residues from the N-terminal side of the full length protein does not alter its stability. Furthermore, normalization of probes shows that the urea-induced denaturation of both the proteins is a two-state process. Folding of both structural variants (Tr-αC-PE and FL-αC-PE) of P. tenue were also studied using molecular dynamics simulations at 300 K. The results show clearly that the stability of the proteins is evenly distributed over the whole structure indicating no significant role of N-terminal residues in the stability of both proteins.

Geitlerinema is a dominant benthic filamentous cyanobacterium found in the Gulf of Thailand and the Andaman Sea. We investigate the diversity of Geitlerinema strains isolated from Thailand using morphological and molecular characterization. No morphological differences were observed in the Geitlerinema isolates. Nucleotide sequencing and phylogenetic analyses of the 16S rDNA, the 16S-23S rRNA internal transcribed spacer (16S-23S ITS) and the cpcB-cpcA intergenic spacer (cpcB-cpcA IGS) showed that the marine Geitlerinema isolates belong to a single cluster that includes marine Geitlerinema sp. Flo1 and Geitlerinema sp. PCC7105 but is distinct from a type species, Geitlerinema splendidum and Geitlerinema strains which were recently transferred to the genus Anagnostidinema. From our results, it is confirmed that marine Geitlerinema differs from true freshwater Geitlerinema and is proposed to be a new genus in Oscillatoriales. In addition, using random amplification of polymorphic DNA (RAPD), the marine Geitlerinema isolates in this study could be classified into eight clades; however, this classification revealed no correlation with the geographic locations.
