Alloferon-1 ameliorates acute inflammatory responses in λ-carrageenan-induced paw edema in mice

Abstract: Alloferon-1 have been proposed as an effective peptide to enhance antitumoral immunity, antiviral defense and anti-inflammatory activity. This work aimed to assess anti-inflammatory effects of alloferon-1 against acute inflammation and histopathological deformations in λ-carrageenan-induced paw edema in mice. Systemic pretreatment with alloferon-1 (22.0 mg/kg) intraperitoneally injected mice showed a significant reduction in paw thickness and vascular permeability. Alloferon-1 prevented λ-carrageenan-evoked ex… Show more

“…In brief, compounds 8 and 9 were capable of decreasing paw edema, which was verified by a prominent percentage of inhibition, such as 44.83% and 49.29% respectively, at the end of the experiment. Understanding the mechanisms of acute inflammatory response is the basic step in proposing novel compounds as anti-inflammatory agents [ 45 ]. As relevant and widely used for testing novel compounds, carrageenan-induced paw edema represents an acute inflammation model characterized by biphasic edema.…”

“…As relevant and widely used for testing novel compounds, carrageenan-induced paw edema represents an acute inflammation model characterized by biphasic edema. During the first hour, there is a release of histamine, serotonin, tachykinin, bradykinin, and other pro-inflammatory agents that immediately cause clinical manifestations such as edema, hyperalgesia, and erythema, appearing immediately [ 45 , 46 ]. This is in accordance with our observations, i.e., the greatest paw edema because of venous obstruction and elevated vascular permeability was noticed in all groups in the first two hours [ 47 ].…”

“…Additionally, in HR-1 hairless mice, topical application of alloferon prevented epithelial thickness induced by UVB irradiation, indicating an inhibitory effect on IL-6 production 119. Furthermore, alloferon has been shown to attenuate acute inflammatory response in λ-carrageenan-induced mouse paw edema in our previous study (Zhang et al, 2022). Administration of the peptide, suppressed the expression of pro-inflammatory cytokines such as TNF-α in inflamed paw tissue, monocyte chemoattractant protein 1 (MCP1), and IL-5 (Zhang et al, 2022).…”

“…The identified amino acid sequences for alloferon are as follows: H-His-Gly-Val-Ser-Gly-His-Gly-Gln-His-Gly-Val-His-Gly-OH (alloferon I) and H-Gly-Val-Ser-Gly-His-Gly-Gln-His-Gly-Val-His-Gly-OH (alloferon II) (Chernysh et al, 2002). Alloferon has displayed anti-inflammatory and antibacterial effects, as well as immunoregulatory and immunomodulatory activities against tumors and viruses within the span of extensive research (Zhang et al, 2022). In this study, we aim to provide a comprehensive review of the biological effects and applications of alloferon and its analogues.…”
