Alliin, a garlic organosulfur compound, ameliorates gut inflammation through MAPK-NF-κB/AP-1/STAT-1 inactivation and PPAR-γ activation

Abstract: Our results show that alliin ameliorates DSS-induced ulcerative colitis and inhibits the inflammatory responses in LPS-stimulated RAW264.7 cells partly through inhibiting ERK1/2-, JNK-/PPAR-γ-stimulated NF-κB/AP-1/STAT-1 activations.

“…Moreover, in dextran sulfate sodium‐induced colitis mouse models, alliin ameliorated ulcerative colitis. Moreover, in in vitro lipopolysaccharide‐stimulated RAW264.7 cell models, alliin also inhibited the inflammatory response, and this effect might be explained by alliin suppressing the MAPK‐PPAR‐γ/NF‐κB/AP‐1/STAT‐1 signaling pathways and ameliorating gut inflammation (Shi et al, ).…”

“…Alliin (S‐allyl‐L‐cysteine sulfoxide) is one of the major sulfur‐containing compounds in garlic (Hirsch et al, ). Alliin occurs in species of the Allium genus, such as onion, leek, and green Chinese onion, in addition to garlic (Shi et al, ). It exists as its reduced form in intact garlic (Trio et al, ).…”

“…At the same time, phosphorylation of ERK1/2 and p38 is inhibited in Ad/PPAR C2C12 cells, concomitant with inhibition of NF-B translocation from cytosol to nucleus [46] . Likewise, Shi and the colleagues demonstrated that alline, a potent PPAR- activator, ameliorates LPS-induced production of iNOS, IL-1, IL-6 and TNF- from RAW264.7 cells through the reduced phosphorylation of ERK1/2, JNK and p38, suggesting that PPAR- activation regulates MAPKs activity [47] . These suggest PPAR activation attenuates inflammatory response through regulation of protein kinase activity (Figure 2c).…”
