ALKBH5 in mouse testicular Sertoli cells regulates Cdh2 mRNA translation to maintain blood–testis barrier integrity

Abstract: Background RNA N6-methyladenosine (m6A) is involved in mammalian spermatogenesis. In both germ cells and Leydig cells, ALKBH5 regulates spermatogenesis and androgen synthesis in an m6A-dependent manner. However, it is unclear whether ALKBH5 plays a role in testicular Sertoli cells, which constitute the blood–testis barrier (BTB) through cell junctions between adjacent Sertoli cells. Methods ALKBH5 expression in the testes of humans and mice was det… Show more

“…The readers of m6A are considered essential to function in modulating gene expression [ 27 , 28 ]. To explore how m6A modification regulates CHST11 expression, the interaction between CHST11 and m6A binding proteins, including YTH domain family (YTHDF) proteins, IGF2BPs, and hnRNP proteins, was analyzed.…”

“…IGF2BP family, which includes IGF2BP1, IGF2BP2, and IGF2BP3, are m6A reader proteins. This family has been proven to increase the stability and translation of mRNA by binding to target mRNAs [ 11 ]. Among these, IGF2BP3 exhibits high expression in various tumor tissues, such as lung cancer [ 12 ], gastric cancer [ 13 ], pancreatic cancer [ 14 ], kidney cancer [ 15 ], hepatocellular carcinoma [ 16 ], breast cancer [ 17 ], and ovarian carcinoma [ 18 ], in contrast to adjacent normal tissues, indicating its role in promoting tumorigenesis [ 19 ].…”
