Alkaloids from Black Pepper (Piper nigrum L.) Exhibit Anti-Inflammatory Activity in Murine Macrophages by Inhibiting Activation of NF-κB Pathway

Abstract: Black pepper (Piper nigrum L.) has been commonly utilized in food preparation and traditional medicine in several countries. Seven new amide alkaloids, pipernigramides A–G (3, 10, 38, and 41–44), a new piperic ester, pipernigrester A (48), along with 47 known compounds were isolated from the EtOH extract of P. nigrum. The inhibitory effects on nitric oxide (NO) of all compounds were then evaluated. Among the tested compounds, three of them (42–44) significantly inhibited inducible nitric oxide synthase (iNOS)-… Show more

“…The isolated alkaloid from P. nigrum exhibited anti-inflammatory effect in RAW 264.7 cells stimulated by LPS and significant inhibition in iNOS-mediated NO and IL-1β, IL-6, and TNF-α. It also demonstrated anti-inflammatory activity in edema induced by carrageenan (Pei et al, 2020). Reports have confirmed the improvement of ovalbumin-induced nasal epithelial barrier dysfunction in allergic rhinitis mouse model.…”

“…Natural resources have attracted considerable interest by many scientists on the natural extracts identification due to their potential applications in treating human health problems and enhancing food security (Li et al, 2014(Li et al, , 2015. Traditional spices and food flavoring agents, such as star anise and black pepper, date back several thousand years (Liu Q. et al, 2020;Pei et al, 2020;Wang et al, 2021). They played important roles in clinical therapy and the food industry due to their beneficial medical use (Moghaddasi et al, 2018;Kamoun et al, 2019;Li H. et al, 2019), strong pharmacological activity (Abukawsar et al, 2018;Yu et al, 2020), antioxidant potential (Liu et al, 2008), and low cost (Destro et al, 2019;Orrillo et al, 2019;Ding et al, 2020).…”

“…Various alkaloids within black pepper have been shown to ameliorate LPS-induced inflammation in mouse models. Levels of LPSinduced pro-inflammatory cytokines IL-1β, TNF-α, and IL-6 were reduced by guineensine (Reynoso-Moreno et al, 2017), piperine (Wang-sheng et al, 2017;Ren and Liang, 2018), and three alkaloids of unknown identity (Pei et al, 2020). Additionally, piperine and the three unknown alkaloids were shown to inhibited LPSstimulated pro-inflammatory Prostaglandin E 2 release (Wangsheng et al, 2017;Pei et al, 2020).…”
