Alkaloid Production From Papaver Tissue Cultures

Abstract: Papaver bracteatum was established as cell suspension culture, as suspension culture with callus grown roots, and as suspension culture containing embryoids and shoots. Alkaloid analysis using thin layer chromatography and reverse phase high pressure liquid chromatography detected the presence of thebaine (0.007%) in shoot cultures. Papaver somniferum cell suspension cultures and suspension cultures containing callus grown roots were established also. Cryptopine was isolated and identified by its mass spectrum. Show more

“…This discrepancy has several different explanations: the use of different analytical methods with varied sensibilities, the use of different Papaver somniferum cultivars [7] and the analysis of somatic tissue cultures at various stages of differentiation. It is known that organogenesis or embryogenesis are accompanied by morphinan alkaloid accumulation [24,25]. We have also established Papaver somniferum tissue cultures showing different degrees of differentiation controlled by various hormonal conditions.…”

“…Although other HPLC methods for the separation of one or several alkaloids of Pupauer spp. have been reported (Hodges and Rapoport, 1982;Kamo et a/., 1982;Staba et al, 1982;Khanna and Shukla, 1986;Roberts ef al., 1987;Tyler et al, 1988), these have often involved buffered mobile phases, two solvent systems and band asymmetry. To our knowledge, a rapid and simple HPLC method for the qualitative and quantitative evaluation of several major alkaloids of P. sorntziferurn and the alkaloid precursor L-tyrosine has not been previously described.…”

“…61 MS/MS spectra from both the TCD-IMS/MS system and Orbitrap identify 370 m/z as cryptopine (C 21 H 23 NO 5 ), as described before. 62 The relationship between opium poppy and the alkaloids N,N-dimethylnarcotine, reticuline, and cryptopine has been previously established and is not the result of chemical interferences in the seized samples. 63 The peak at 268 m/z occurs as a loss of water from morphine, 55 and water loss from codeine accounts for the ion at 282 m/z.…”
