Alisol A 24-Acetate and Alisol B 23-Acetate Induced Autophagy Mediates Apoptosis and Nephrotoxicity in Human Renal Proximal Tubular Cells

Abstract: Two natural compounds alisol A 24-acetate (24A) and alisol B 23-acetate (23B) are abundant in Rhizoma alismatis. In the present study, we evaluated the induction of 24A and 23B on apoptosis and possible nephrotoxicity of human renal proximal tubular (HK-2) cells by activating autophagy and also explored its regulation on PI3K/Akt/mTOR signaling pathway. Presently, Clusterin, Kim-1, and TFF-3 were considered to be new bioindicators of nephrotoxicity. Interestingly, the protein expression and mRNA levels of Clus… Show more

“…Juzepzuk (Alismataceae), is a natural medicine widely cultivated in China, Japan, Korea, India, and Europe. Traditionally, AR is known to exhibit diuretic, hyperlipidemic, inflammatory, antitumoral, and damp-heat clearing actions [ 1 , 2 , 3 , 4 , 5 , 6 , 7 ] and has been used for treating dysuria, edema, urinary tract infection, retention of fluid and phlegm, and vertigo for more than 1000 years [ 8 ]. As a folk medicine that has long been used to promote health and longevity, AR is now included in the Pharmacopoeia of China as a representative traditional Chinese herbal diuretic.…”

“…(Alismataceae), which is an aquatic plant that has been cultivated mainly in oriental countries (e.g., China, Japan, and Korea) but is also widely distributed in other areas (e.g., North America and Europe). AR has various activities, such as diuretic, hyperlipidemic, inflammatory, antitumoral, and damp-heat clearing activities [ 2 8 ]. AR is traditional folk medicine that has long been used to promote health and longevity (Sheng Nong's herbal classic) for more than several thousand years.…”

“…It is commonly used to treat di erent diseases, including cancer and induced autophagy-dependent cell death through the accumulation of intracellular ROS and subsequent activation of JNK signaling pathway in human colon cancer (Zhao et al 2017). In human renal proximal tubular cells, AB23A-induced cell apoptosis is mediated by autophagy and nephrotoxicity via PI3K/AKT/mTOR signaling pathway (Wang et al 2017). erefore, more and more attention has been paid to AB23A because of its potential anti-cancer e ect and the research value in the treatment of human cancer.…”
