Alginate-Based Platforms for Cancer-Targeted Drug Delivery

Abstract: As an acidic, ocean colloid polysaccharide, alginate is both a biopolymer and a polyelectrolyte that is considered to be biocompatible, nontoxic, nonimmunogenic, and biodegradable. A significant number of studies have confirmed the potential use of alginate-based platforms as effective vehicles for drug delivery for cancer-targeted treatment. In this review, the focus is on the formation of alginate-based cancer-targeted delivery systems. Specifically, some general chemical and physical properties of alginate … Show more

“…In cancer research, alginate-based nanomaterials emerged based on their potential to reduce the side effects of anti-cancer chemotherapeutic agents and enhance their efficacy in targeting and penetrating the target cells. They can also be used to overcome the chemotherapeutic resistance, which could result from several intracellular pathways of drug uptake and a higher accumulation of drug inside cancer cells ( Alfarouk et al, 2015 ; He et al, 2020 ). For instance, Katuwavila et al have encapsulated doxorubicin (DOX) in chitosan-alginate NPs, which were cross-linked through the ionic gelation method.…”

“…Alginate-based nanomaterials are among the most extensively characterized biopolymers used to develop targeted or localized delivery systems for therapeutic compounds, including bee products or their extracts ( Abasalizadeh et al, 2020 ; Homem et al, 2021 ; Saberian et al, 2021 ). This is due to their offering desirable characteristics that include a high therapeutic-payload, targeted efficiency, pH sensitivity, capability of protection from degradation, thickening properties, gelling abilities, high availability, and relatively low cost ( Lee and Mooney, 2012 ; Sun and Tan, 2013 ; He et al, 2020 ).…”

“…[65][66][67] Additionally, a variety of drug delivery systems (DDS) based on alginate, including microcapsules, microparticles, gel particles, pellets, and beads, have been developed as well and proved to be able to promote drug stabilization and longer release. [68,69] Alginate-based materials offer also great promises in the targeted treatment of cancer [70,71] and the development of specific biosensors. [72,73] A summary of the most common uses of alginate is reported in Figure 2.…”

“…In this sense, among the broad variety of materials that can be used for this purpose, alginate, due to its unique physicalchemical and biological properties, has shown relevant promises in the development of highly effective chemotherapy nanocarriers. [71,177,178] By way of example, Alg-NPs loaded with curcumin and resveratrol were investigated for the treatment of prostate cancer showing marked cytotoxicity effect on tumor cells without producing hemolysis, hence being safe for intravenous administration. [179] Similarly, oxidized alginate NPs conjugated with doxorubicin and containing curcumin demonstrated lower toxicity toward health cells compared to analogous free drug formulations and pH-responsive delivery for the targeted treatment of breast cancer.…”
