Alfalfa Mosaic Virus Replicase Proteins P1 and P2 Interact and Colocalize at the Vacuolar Membrane

Abstract: Replication of Alfalfa mosaic virus (AMV) RNAs depends on the virus-encoded proteins P1 and P2. P1contains methyltransferase-and helicase-like domains, and P2 contains a polymerase-like domain. Coimmunoprecipitation experiments revealed an interaction between in vitro translated-P1 and P2 and showed that these proteins are present together in fractions with RNA-dependent RNA polymerase activity. A deletion analysis in the yeast two-hybrid system showed that in P1 the C-terminal sequence of 509 amino acids with… Show more

“…BMV 1a and 2a proteins and newly synthesized RNAs also colocalized with electron-dense bodies widespread in the cytoplasm and probably of the same origin in barley leaf sections after detection by IGL (7). AMV minus-strand RNA and the replication-associated P1 and P2 proteins were detected in association with the tonoplast in cowpea protoplasts by confocal microscopy, although P2 was found both at the tonoplast and in the cytoplasm (34). Hence, AMV and CMV replicase proteins colocalize at the same subcellular compartment, a feature possibly shared by the ilarvirus Tobacco streak virus (18).…”

“…The sites of accumulation of the replication-associated proteins and minus-strand RNA have been identified for two other members of the family Bromoviridae, the bromovirus BMV and the alfamovirus AMV (7,21,34). BMV minusstrand RNA and the replication-associated proteins 1a and 2a colocalized with vesicles originating from the ER, mostly concentrated in the perinuclear region in infected barley protoplasts, as analyzed by immunofluorescence confocal microscopy (21).…”

“…3B, lanes 13 and 14). We note, however, that the CP/ RNA ratios are different between the two assays and that the viral polymerase preparation is naturally contaminated with CP (22,26), which would result in even higher CP/RNA ratios. Nevertheless, comparison of pk1/hp2 with pk3/hp2 shows that a stronger pseudoknot is capable of preventing CP of inhibiting minus-strand synthesis at a CP/RNA ratio of 0.5 to 1.…”

“…Samples from the P30 pellets corresponding to 50 mg of infiltrated tissue were loaded onto a Western blot. P1 and P2 were detected using antisera to the C-terminal amino acids 1100 to 1120 of P1 and the N terminus of P2 (36,38). Western blotting was performed with Hybond-P polyvinylidene difluoride membranes (Amersham Pharmacia Biotech).…”
