Aldehyde Dehydrogenases: Measurement of Activities and Protein Levels

Abstract: Seventeen enzymes are currently viewed as belonging to the human aldehyde dehydrogenase superfamily, and all of them catalyze the pyridine nucleotide-dependent oxidation of aldehydes to acids. Depending on the specific aldehyde dehydrogenase, lack of sufficient catalytic activity (1) results in a gross pathological phenotype in the absence of any insult, or (2) is ordinarily of no consequence with respect to gross phenotype, but is of consequence when the organism is subjected to a relevant insult. Described i… Show more

“…B, proposed mechanism for ␤-elimination of Aldi-1, which yields two products, Aldi-1v and Aldi-1n. specifically, ALDH1A1 and ALDH3A1 activity was suggested as a predictor of efficacy against cyclophosphamide treatment in breast adenocarcinoma (34). Reduction of ALDH activity increases the sensitivity of the human lung adenocarcinoma cell line A549 to a metabolite of cyclophosphamide, 4-hydroxyperoxycyclophosphamide (5).…”

“…It was originally designated as the tumor ALDH as it was found highly expressed in some human tumors such as hepatoma, lung adenocarcinoma, myeloma, breast cancer and stem cell populations. [20,21,22,23] It is also expressed in some normal human tissues such as cornea and keratinocytes. [18,24] Despite being present in normal cells, studies have shown that Aldh3a1(-/-) knockout mice are viable.…”

“…GSTM1 and GSTT1 have been shown to conjugate isothiocyanates, and in turn isothiocyanates have been shown to induce GSTs. Induction in enzyme activity occurs rapidly within several days of cruciferous vegetable consumption and declines with removal of the vegetables from the diet [72]. Polymorphisms in the GSTM1 and GSTT1 genes result in complete lack of GSTM1-1 and GSTT1-1 proteins, respectively.…”
