Alcohol and nicotine polygenic scores are associated with the development of alcohol and nicotine use problems from adolescence to young adulthood

Abstract: Background and Aims Molecular genetic studies of alcohol and nicotine use have identified many genome‐wide association study (GWAS) loci. We measured associations between drinking and smoking polygenic scores (PGS) and trajectories of alcohol and nicotine use outcomes from late childhood to early adulthood, substance‐specific versus broader‐liability PGS effects, and if PGS performance varied for consumption versus problematic substance use. Design, setting, participants and measurements We fitted latent growt… Show more

“…Polygenic scores have been used in research for disease prediction and to evaluate disease correlations, with potential for clinical translation to identify high-risk individuals 6 . In particular, tobacco use behaviors have shown genetic correlations with diseases such as schizophrenia and substance use disorders [7][8][9][10][11] .…”

“…More recently, polygenic scores (PGS), which aggregate risk for a trait across the genome using information from genome-wide association studies (GWAS), were robustly associated with substance use 12 and substance related problems 13 across adolescence and into young adulthood. However, though robustly associated, current PGS do poorly in identifying individuals affected by SUDs 14 .…”
