Alantolactone suppresses inflammation, apoptosis and oxidative stress in cigarette smoke-induced human bronchial epithelial cells through activation of Nrf2/HO-1 and inhibition of the NF-κB pathways

Abstract: Background: It is well established that airway remodeling and inflammation are characteristics for chronic obstructive pulmonary disease (COPD). Moreover, cigarette smoke extract (CSE) promots inflammation, apoptosis and oxidative stress in COPD. And, there is evidence suggested that alantolactone (ALT), a sesquiterpene lactone isolated from Inula helenium, plays an adverse role in inflammation, apoptosis and oxidative stress. However, few studies have investigated the function and mechanism of ALT treatment o… Show more

“…In this experiment, dioscin inhibited the production of ROS by activating AMPK-Nrf2 in mastitis mice and mMECs. Recent studies exemplified that Nrf2 activation can inhibit inflammation-mediated damage through its antioxidant effect [ 55 ]. However, the effects of Nrf2 on the NF- κ B signaling pathway, NLRP3 inflammasome, and cell pyroptosis remain controversial [ 56 , 57 ].…”

“…Another study investigated the function and mechanism of ALT treatment on COPD pathological process. Thereby, protective effects of ALT against inflammation, apoptosis and oxidative stress in cigarette smoke-induced human bronchial epithelial cells through activation of Nrf2/HO-1 and inhibition of the NF-κB pathways could be shown [ 200 ].…”

“…The activation of the Nrf2/HO-1 signaling pathway by onopordopicrin is a characteristic shared by other sesquiterpene lactones. For instance, the two sesquiterpene lactones hemistepsin A from Hemistepta lyrata and alantolactone from Inula helenium have been shown to activate the Nrf2/HO-1 pathway and to protect against oxidative stress in human keratinocytes and bronchial epithelial cells, respectively [ 45 , 58 ]. Another sesquiterpene lactone, parthenolide, found in feverfew ( Tanacetum parthenium ) and belonging, like onopordopicrin, to the germacrane subclass, was shown to have antioxidant properties by increasing the expressions levels of Nrf2 and HO-1 in H 2 O 2 -treated osteoblasts [ 59 ].…”
