Macroalgae that grow in the tropics are continuously exposed to ultraviolet radiation and sunlight. Macroalgae have self-defense to avoid damage caused by ultraviolet radiation by carrying out secondary metabolism and producing several bioactive compounds. The bioactive compounds produced have the potential to be utilized to inhibit free radicals that can cause degenerative diseases in humans. This research aims to identify bioactive compounds and determine the antioxidant activity, total phenolic compound, total flavonoid compound, and pigment content of red algae Gracilaria sp. from Bintan island. This research has the novelty of the sampling location to be later developed into a sustainable resource in the pharmaceutical field. Gracilaria sp. found in Bintan island was taken, dried in indirect sunlight, and macerated using universal solvent methanol. Phytochemical testing was conducted to determine the presence of flavonoids, steroids, triterpenoids, saponins, and tannins. Antioxidant activity was determined using DPPH as a free radical at the maximum wavelength of DPPH (515 nm) with an incubation time of 55 minutes. Total phenolic and flavonoid compounds were determined using standard solutions of gallic acid and quercetin, respectively. The results showed that the methanol extract of Gracilaria sp. contains flavonoid and tannin compounds, has a chlorophyll-a content of 0.43 mg/g, a carotenoid content of 0.91 µmol/g, the total phenolic compound of 4.67 mg/GAE g, total flavonoid compound of 2.26 mg/QE g and has antioxidant activity with IC50 value of 982.25 ppm. Utilization of Gracilaria sp extract from Bintan waters as a pharmaceutical ingredient is expected to increase prospects and sustainable use in the pharmaceutical and health fields.

Background and Aim: Swamp buffaloes play an important role in the rural economy of Indonesia. They consume various forages during their grazing time, including those with anti-parasitic potential. However, the information about the type and quality of forages and their potential as a natural anthelmintic for swamp buffalo is very limited. This study aimed to identify the diversity, quality, and anthelmintic potency of forages consumed by swamp buffaloes reared in Bantarkawung Subdistrict, Brebes District, Central Java Province, Indonesia. Materials and Methods: Samples of forages were obtained during three observation periods of the study, with a 12-week interval between each period. Forage diversity was evaluated by identifying its consumption by swamp buffaloes during their grazing activity in the field and feeding time in their shed. The quality of forages was analyzed using proximate analysis to measure their dry matter (DM), crude fiber (CF), crude protein (CP), crude fat (CFat), and ash contents. This is followed by the calculation of their total digestible nutrient based on the proximate analysis results. Botanical composition analysis was then conducted to measure the predominance of forages consumed by the livestock during their grazing activity. Literature reviews were carried out to explore forage’s anthelmintic activity. Results: The results showed that swamp buffaloes consume nine species of forage in the shed and 47 in the grazing area, including nine legumes, 18 grass, and 20 others. Swamp buffaloes consumed forages of lower quality, which contained high CF contents and varying levels of other nutrients below their daily nutritional needs. The grazing activity allowed swamp buffaloes to consume a higher variety of forages with better nutritional quality, thereby enabling them to meet their nutritional needs. Legumes and other forages served as the major protein sources, providing CP of 20.03% DM and 11.53% DM, and CF levels of 17.01% DM and 20.35% DM, respectively. The results also showed that the consumption of these forages increased during the rainy season. The predominant species of legumes consumed were Leucaena leucocephala and Acacia spp., while Alternanthera sessilis and Merremia umbellata were the predominant species of other forages. A total of 13 of the 47 species could potentially be used as natural anthelmintic due to their secondary metabolites, namely, tannin, flavonoid, saponin, terpenoid, diterpenoid, and mimosine. These compounds exert anthelmintic effects by inhibiting egg-hatching and larval development, as well as damaging the surface structure of both larvae and adult worms, ultimately leading to the death of the parasites. Conclusion: Overall, swamp buffaloes consumed more variety of forages during grazing compared to when they were kept in sheds. While the low-variety and low-quality forages provisioned for swamp buffaloes in their shed resulted in a low nutrient intake below their daily requirement. Furthermore, daily grazing activities allowed swamp buffaloes to fulfill and supplement their need by consuming a variety of grasses, legumes, and other forages in their respective grazing areas. Some of these forages also have the potential to become natural anthelmintic because they contain secondary metabolites, such as tannins, flavonoids, saponins, terpenoids, diterpenoids, and mimosine. Sargassum sp merupakan jenis rumput laut yang tumbuh subur secara alami di perairan Indonesia, termasuk di Perairan Simeulue, Provinsi Aceh. Sargassum sp. mengandung berbagai senyawa bioaktif yaitu fenol yang berpotensi sebagai antioksidan. Sargassum sp. di Perairan Simeulue belum dimanfaatkan oleh masyarakat setempat dan belum ada laporan ilmiah tentang total fenol dan aktivitas antioksidan dari rumput laut tersebut. Tujuan penelitian ini menentukan aktivitas antioksidan dan total fenol Sargassum sp yang terdapat di Perairan Simeulue. Metode penelitian meliputi pengambilan sampel rumput laut, preparasi dan ekstraksi sampel rumput laut, uji fitokimia, analisis total fenol dan analisis aktivitas antioksidan rumput laut. Analisis data menggunakan analisis regresi linear untuk melihat hubungan total fenol dengan aktivitas antioksidan DPPH Sargassum. Hasil penelitian menunjukkan bahwa total fenol dan aktivitas antioksidan lima jenis rumput laut genus Sargassum (S. muticum, S. binderi, S. crassifolium, S. granuliferum, S. fluitans) di Perairan Simeulue menunjukkan nilai yang bervariasi. Total fenol 7,45-9,02 mg GAE/g dengan nilai tertinggi terdapat pada S. binderi dan nilai terendah pada S. fluitans. Aktivitas antioksidan 74,7-152,4 μg/mL dengan nilai tertinggi terdapat pada S. binderi, sedangkan nilai terendah terdapat pada S. fluitans. Hasil analisis korelasi (R2 = 0,9059) menunjukkan bahwa aktivitas antioksidan rumput laut genus Sargassum berhubungan erat dengan total fenol. Kelima spesies rumput laut Sargassum di Perairan Simeulue mempunyai aktivitas antioksidan yang kuat (74,7-152,4 μg/mL) sehingga berpotensi dikembangkan sebagai antioksidan.
